Objective: Endothelin-1 was found by Japanese scholar Yanagisawa in 1988,it was first taken from vascular endothelial cells of pig .It is a peptide composed by 21 Amin Acids with many biological effects. The research about the relation of ET-1 and neoplasms has been focused. This study detects the expression of ET-1 in lung cancer and in circum-lymph node metastases, discusses the relationship between the ET-1 positive expression and the histological subtype\cell differentiation\TNM staging\tumor size and lymph node-metastasis. This study demonstrates the role of ET-1 in the generation\development and metastasis of lung cancer Methods: The ET-1 express in the 80 lung cancer tissues and the circum-lymph nodes were detected by the avidin-bioctin-peroxidase complex immunohistochemical method. The pathologic studies on carcinoma of the lung cancer established by the standard of WHO (1981) Result: In all the 80 tumor tissues sections, 45 cases positive expressed ET-1.In the 50 circum-lymph node raetastases, 29 cases positive expressed ET-1 .The ET' s positive rate of lung adenocarcinoma was 62. 5%(21/34), while the ET' s positive rate of lung squamous-cell carcinoma was 68.7%(22/32), The ET' s positive rate oflung small -cell-lung cancer was 15.0%(2/14). The ET' s positive rate of SCLC was 15.0% while NSCLC was 65.2%, there was statistical difference between them. ( x 2=12.14 p<0.05) For TNM stage I -III tumors , The ET' s positive rate of I was!9. 0%, while II was 65.7% and III was75.0%, there was statistical difference between them.. The ET' s positive rate of tumor size for diameter>3cm was 65.6%, while diameter<3cm was 36.3%, there was statistical difference between them. The rate of circum-lymph nodes metastasized with ET-lpositive express in tumor was 79.1%, it is higher than that with ET-1 negative express (47.8%). ( x 2=6. 74 p<0.05). In adenocarcinoma and squamous-cel1 carcinoma, The ET' s positive rate of expression in moderate and well differentiated types was 71.9%, while the poorly differentiated type was 58.8%, there was no statistical difference between them. ( x2=1.24 p>0. 05) Conclusion: l.ET-1 lies in the cells of lung cancer and in the circum-lymph node-metastases.2. The positive expression of ET-1 in lung cancer is correlated with histological subtype\tumor size\pathologic stage and lymph node involvement. But there is no significant association between ET' s expression and patient' s age and differentiation grade.3. ET-1 may be used as a valuable biomarker that plays an important role in the response of lung adenocarcinoma and squamous-cell carcinoma to grow and metastasize. Itis a useful predictor of prognosis. It is helpful toprovide new therapy assistant methods.
