Objective:The expression of ERα/β, COX-2, TGFβ1 protein was detected with immunohistochemical method in normal breast tissues, intraductal papilloma (disease), carcinoma in situ, invasive cancer tissues and analyzed. Our aim was to explore their expression changes and significance in the development of breast cancer.Methods:Immunohistochemical (SP) method was used to detect the expression of ER-α/β, COX-2, TGF-β1 protein in.breast normal tissue group (61), intraductal papilloma (disease) (65), carcinoma in situ (45), invasive breast cancer group (87) and analyzed.Results:1. the expression of Era and ErβThe overall expression level of ERa protein in papilloma, carcinoma in situ, invasive cancer tissues was differences statistically.in compared with normal epithelium(P<0.01). The expression scope of ERa protein in papilloma, carcinoma in situ, invasive cancer tissues was differences statistically.in compared with normal (P<0.01).But there was no differences statistically among them. There was differences statistically between papilloma and normal (P<0.05), the expression of carcinoma in situ and invasive cancer tissues was differences statistically.in compared with normal (P<0.01), the expression of carcinoma in situ and invasive cancer tissues was differences statistically.in compared with papilloma (P<0.05)The expression level of Erβprotein in normal epithelium, papilloma, carcinoma in situ, invasive cancer tissues was high,there was differences statistically between invasive cancer tissues and normal hyperplasia breast tissue (P<0.05); the expression scope of Eβin the breast cell was same, there was differences statistically between papilloma and normal tissue (P< 0.05).but compared to the expression in the cell,,there was differences statistically between invasive cancer tissues and normal hyperplasia breast tissue.The expression scope of ERa/Erp in the normal breast epithelium, papilloma, carcinoma in situ, invasive cancer tissues was no differences.The overall expression and expression intension was increase gradually accompany with the development of disease Severity, especially the expression of ERa/Erβin the normal breast epithelium (0.34), papilloma (0.43), carcinoma in situ (0.68), invasive cancer tissues (0.71) was increase.2. the expression of COX-2 proteinThere was highly expression of COX-2 protein in the normal epithelium, papilloma, carcinoma in situ, invasive cancer tissues. The overall expression of COX-2 in papilloma, carcinoma in situ, invasive cancer tissues was differences statistically in compared with the normal breast epithelium (P<0.01). The expression of COX-2 in papilloma and carcinoma in situ was differences statistically in compared with invasive cancer tissues(P<0.05).There was differences statistically between carcinoma in situ and invasive cancer tissues (P<0.01).But there was no differences statistically among them (P> 0.05). The expression intensity of COX-2 in invasive cancer tissues was differences statistically in compared with the normal epithelium (P< 0.05). The expression intensity of COX-2 in papilloma and carcinoma in situ was differences statistically in compared with the normal epithelium (P<0.01). The expression intensity of COX-2 in papilloma and carcinoma in situ was higher than in invasive cancer tissues (P<0.01)3. the expression of TGFβ1 proteinThe overall expression level of TGFβ1 protein in papilloma, carcinoma in situ, invasive cancer tissues was differences statistically.in compared with normal epithelium (P<0.01),there was differences statistically among papilloma, carcinoma in situ and invasive cancer tissues(P<0.05).The expression scope in normal epithelium, papilloma, carcinoma in situ and invasive cancer tissues was wide. The expression in papilloma was lower than in normal epithelium (P<0.05). The expression in carcinoma in situ and invasive cancer tissues was higher than in normal epithelium (P<0.01). The expression intensity of TGFβ1 protein between in papilloma and in normal epithelium was no differences statistically (P>0.05).The expression intensity in carcinoma in situ and invasive cancer tissues was higher than in papilloma and normal epithelium(P<0.05).But there was no differences statistically between in carcinoma in situ and invasive cancer tissues(P>0.05).Conclusion:1. The expression intensity of ERβin normal epithelium, papilloma, carcinoma in situ, invasive carcinoma were gradually decreased, the expression of ERa and ERβduring the development of breast cancer are the contrary, suggesting that ERβmay be a self-protective factors of breast cancer.It may become a new target for gene therapy of breast cancer and provide the basis for early diagnosis and prognosis.2. The positive rate of TGFβ1 expression in the normal group, papilloma, carcinoma in situ and invasive carcinoma group was gradually increased, suggesting that TGFβ1 may promote the development of breast cancer, it was significant for early diagnosis and prognosis of breast cancer.3. The positive rate of COX-2 protein expression in Papilloma group was significantly higher than in the normal group, carcinoma in situ and invasive carcinoma groups, suggesting that COX-2 protein expression may be an early event in breast cancer, and the detection of COX-2 may be contributed to early clinical diagnosis...
