THE INTERVENING EFFECT OF UROKINASE ON ICR MICE?S MODS Objective: To investigate the role of the local microthrombi formed in the organ, in the pathogenesis of MODS and the intervening effect of clearing up the inicrothrombi and mending the microcireulation, by means of intramuscularly injecting urokinase, on ICR mice?s MODS. Methods: 120 male ICR mice, average 20. 40?. 05g per animal, were randomized into control group (group C, 55 animals) , experiment group (group E, 55 animals), normal group(group N, 5 animals) and urokinase group (group Mi, 5 animals). In order to prepare the animal model of MODS, both the group C and group E were given an aseptic intraperitoneai injection with Zymosan A suspended at 25mg/mi, in paraffin oil. The dose was ling/g body weight. After preparing the animal model, administrated the group E urokinase lOmg/g body weight/day intramuscularly injection from the beginning of the experiment (DO), and the group C was not gave any treatment. The 5 animals of group N were absolute normal. The definition of group Ml was that the normal animals which were administrated the same urokinase of group E. Observe and compare the changing of the following indeses: the curve of average body weights dead rate of the first phase and the third phase, relative weight of organ (wet organ weight/body weight Xl 00%), the plasma level of TNF?a (RIA), IL?(ABC桬LISA) and the pathological examination (microscope, electron microscope) of the lung, the liver and the kidney at the 1st day(D1), the 5th day(D5) and the 9th day(D9) of the experiment. Results: The curves of average body weight of 5 group E and group C were almost one curve at the first and the second phase, but from D7, they began to separate. The curve of group E keep increasing, and it of the group C keep decreasing. At D10, the curve of group C increased suddenly and went beyond it of group E. The dead rate of the first phase of group C and group E was 50% and 40% respectively. The difference wasn?t significant (P~O. 751) and it of the third phase was 90% and 33% respectively.
