Fibrin tissue adhesive is a kind of composite composed of several human plasma components. Because of its good stickiness, biogradability, biocompatibility and ability of improving tissue regeneration, it has been utilized more extensively than ever. ~The purpose of the present study is to evaluate the effects of FG on the proliferation and alkaline phosphatase activity of human PDLCs and periodontal tissue regeneration by employing cell culture in vitro and animal experimental technique in vivo. 1. The effects of fibrin tissue adhesive on the proliferation and alkaline phosphatase activity of periodontal ligament cell Using cell culture technique. MTT and enzyme kinetics methods. Results show, the proliferation and ALPase activity of PDLCs in experimental groups are significantly increased in acertain range, and indicate FG can improve the proliferation and ALPase activity of of human PDLCs. 2. The effect of fibrin tissue adhesive to the adhesive rates of PDLCs PDLCs are observed on the FG , to investigate their adhesive rates on FG. Put the Ri on the floor of the culture plates well-distributed, inoculate PDLCs on the FG, then assess the adhesive rates on FG at different time. The adhesive rates on FG reach the max at the 4~ hour after inoculation, whereas, the ones of e-PTFE membrane groups and control groups reach max at 1 O~ and 12 ~?hour respectively. The result indicate that FG can improve the adhesive rate of PDLCs. 3. The effects of fibrin tissue adhesive on the periodontal regeneration. The furcation defects on the premolars of dogs were produced surgically. The defects were devided into 4 equal groups(e-PTFE, FG, e-PTFE combined with FG, control). The results were examined histologically 1 month and 3 ~4i~ ~57~ with FG, control). The results were examined histologically 1 month and 3 months respectively. In experimental groups the JE downgrowth was inhabited in the early healing, and the new attachment formation was promoted while comparing with controls. And there is no significant differences between two experimental groups. Take ePTFE membranes out 6 weeks after operation, observe the PDLCs and bacteria on ePTFE membranes in SEM. The results show that there are more PDLCs ,less bacteria and no epithelial cell on the membrane of the group applied with FG combined with ePTFE membrane than that of the group applied with ePTFE membrane alone. FG has good biocompatibility and can close the primary wound with reducing the downgrowth of junctional epithelium and can significantly improve the periodontal tissue regeneration in treatment of flircation artifieial defects. Especially FG combined with ePTFE membrane can improve the periodontal tissue regeneration significantly in the early stage.
