PPARαis a member of the nuclear receptor super family which has been proposed to play key roles in lipid and carbohydrate metabolism.This study applies PCR-SSCP, CRS-PCR-RFLP, DNA sequencing and PCR-RFLP techniques to detect the polymorphism of PPAR gene from 849 chickens of Gushi-Anak F2 Resource Population.The results were as follows:1. Applying PCR-SSCP method to scan the SNPs, an A/T non-synonymous mutation in the P1 amplified fragment(g.A23884T)was detected by restriction enzyme BglII, resulting in two genotypes that named AA genotype and AB genotype; a G/A non-synonymous mutation in the P3 amplified fragment(g.G29201A)was detected by restriction enzyme BsiEI, resulting in two genotypes that named CC genotype and CD genotype; a T/C silent mutation in the P4 amplified fragment(g.T32311C)was detected by restriction enzyme BspHI, resulting in three genotypes that named EE genotype, EF genotype and FF genotype.2. Estimating the correlation analysis between the polymorphisms and carcass trait,the result showed that different genotypes of P1 locus on the liver weight and liver weight rate were significantly different (P<0.05);different genotypes of P3 locus on the pancreas weight rate was significantly different (P<0.05); different genotypes of P4 locus had effects on the gizzard weight and liver weight rate ,the gizzard weight of FF genotype was higher than EF genotype (P<0.01),comparing with EE genotype, the liver weight rate between EF and FF genotypes were significantly different (P<0.05).3. Estimating the correlation analysis between the polymorphisms and fat, meat quality traits, the result showed that different genotypes of P1 locus on the abdominal fat weight, abdominal fat rate and water loss rate of breast muscle were significantly different (P<0.05); different genotypes of P4 locus had effects on the abdominal fat weight and abdominal fat rate, the differences were very significant (P<0.01), the abdominal fat weight and abdominal fat rate of EE and EF genotypes were higher than FF genotype (P<0.05).4. Estimating the correlation analysis between the polymorphisms and growth trait, the result showed that the different genotypes of P3 locus influenced the birth weight and body weight at the age of 2 weeks very significantly (P<0.01),the shank girth at the age of 8 and 12weeks, pelvis breadth at the age of 8 weeks of CC genotype greater than CD genotype, the difference was significant (P<0.05),the breast angle of CD genotype greater than CC genotype at the age of 4 weeks (P<0.05), EE,EF and FF genotypes of P4 locus had different effects on body weight at the age of 6 weeks,body slanting length at the age of 4 weeks (P<0.01) and body weight,shank girth,pelvis breadth at the age of 4 weeks,length of breastbone at the age of 4,8 and12 weeks (P<0.05).5. Estimating the correlation analysis between the polymorphisms and physiological and biochemical index, the result showed that different genotypes of P1 locus on the total protein and creatinine were significantly different (P<0.05); compared with FF genotype, EE, EF genotypes of P4 site had effects on high density lipoprotein (P<0.05).6. Estimating the correlation analysis between com-genotypes of PPARαgene and economical traits,the result showed that different com-genotypes had significant effects on the liver weight, the pancreas weight rate of carcass trait; different com-genotypes had extremely significant effects on the birth weight and body weight at the age of 6 weeks(P<0.01)and significant effects on body weight at the age of 4weeks, the shank girth at the age of 8 and 12weeks, length of breastbone at the age of 12 weeks(P<0.05) of growth trait;different com-genotypes had significant effects on the abdominal fat weight, abdominal fat rate and water loss rate of breast muscle of fat and meat quality traits; the difference of albumin content between ABCCEE and AACDEF was very significant(P<0.01).
