| Sophora moorcroftiana, a perennial shrub, which belongs to Leguminosae, Snphora L., is one of the endemic species both in Tibet and China. It is produced in the Brahmaputra River valley and Bomi, Nyingchi, Millington, etc. in Tibet and grows in the hillside scrub, sandy floodplain and rocky dry slopes on the height of 2800-4400m. Sophora moorcroftiana is a good eco-plant which is highly resisting of drought, barren, coldness, sand and other anti-ecological adaptation and its main active principle is matrine alkaloids which have high medicinal value which is mainly used for hepatitis B, cirrhosis, colitis diarrhea, gynecological inflammation, pain, swelling and coma, and other diseases treatment, and it is also an important raw material for plant pesticides.In this paper, we used the wild Sophora moorcroftiana as materials to study the content and diversities of alkaloids of different organs, different picking times and different altitudes, and to study the temporal and spatial variation of active ingredients to provide scientific basis for further development. We sampled along the growth zone of Sophora moorcroftiana on the height of 2800-4400m, using acid extraction- alkaline precipitation method for alkaloids extraction and silica gel column chromatography to pure and HPLC to determine the content of total alkaloids, using the oxymatrine content as the index. The main results are as follows:1. Study on the extraction condition of total alkaloids from Sophora moorcroftianaThe results of orthogonal tests show that the best extraction condition is: adding 10-fold amount of 0.5% hydrochloric acid and the extraction time is 20h and extracted thrice.2. The diversity of content of active principle of different apparatus of Sophora moorcroftianaThe active principle of the root is stilbene polymer, while it is matrine alkaloids of the stem, leaf, flower and seed. The alkaloid content in the seeds is the highest, while the stems, leaves and flowers are in very low alkaloid content, which is identical with the traditional Tibetan medicine selection,therefore we used the seeds of Sophora moorcroftiana to study the spatial and temporal variation of alkaloids in this paper.3. The diversity of content of oxymatrine of different sampling times of the seeds of Sophora moorcroftianaSophora moorcroftiana blooms in May-June, going into the fruiting period in September and the fruit maturation is in October. In the experiment we began collecting seeds from the August to research the diversity of the alkaloid content of seeds in August, September, October and winter months. The results showed that the content of oxymatrine increased gradually over time.4. The diversity of content of oxymatrine of different altitudes of the seeds of Sophora moorcroftianaSophora moorcroftiana grows in the hillside scrub, sandy floodplain and rocky dry slopes on the height of 2800-4400m.With the increase of altitudes, sunshine, rainfall and other factors will change correspondingly. With the increase of altitudes, the diurnal temperature difference became larger, which can make the growth cycle become longer, and it is conducive to the accumulation of alkaloids. Overall, with the elevation of altitudes, the content of alkaloids of the seeds of Sophora moorcroftiana present in an increasing trend. |
