The plant polysaccharides were natural macromolecules. People paid considerable attention to them, because of its wide sources, exact effects and natural. Many studies had demonstrated that the plant polysaccharides had a wide range of biological activity. They could enhance immune function, stimulate proliferation of Lymphocyte. The immunity had very important relationship with the free radicals, It could get rid of reactive oxygen species to enhance immunity by Improving the antioxidant. The water soluble alfalfa polysaccharide (WSAP) was extracted from Medicago Sativa L, and the astragalus polysaccharide (APS) was extracted from Astragalus membranaceus. At present, the research of WSAP and APS mainly focused on immunization. The chemical composition and structure of same polysaccharide were differenrt because of different plant origin, different extraction and so on. So the antioxidant of same polysaccharide was differenrt, although the antioxidant of WSAP and APS had reported. The experiments were conducted to evaluate the effects of WSAP and APS on the antioxidative of Abore Acre broilers, and we could provid the theoretical basis for the research on mechanism of WSAP and APS to enhance immunity and the application of animal production.Exp.one The study the content and antioxidant activity of WSAP and APS in vitroIn order to study the content and antioxidant activity of WSAP and APS in vitro, the content of WSAP and APS were determined by anthrone-sulfuric acid method, Free radical-scavenging activities of WSAP and APS were analyzed using free radical production in vitro systems (1,1-dipheyl-2-pic-rylhydrazyl radical ,hydroxyl radical and superoxide anion free radical). The results showed that WSAP and APS could scavenge free radical. The scavenging capacities of the polysaccharides against 1, 1-dipheyl-2-pic-rylhydrazyl, hydroxyl radical and superoxide anion free radical increased with the increasing of their concentrations. The concentrations of WSAP and APS which were necessary to decrease the initial concentration (IC50) of DPPH by 50% were 0.251mg/mL, 0.851mg/mL respectively. The IC50 of WSAP and APS for hydroxyl radical were 0.573mg/mL, 0.907mg/mL. The IC50 of WSAP and APS for superoxide anion free radical were 0.482mg/mL, 0.637mg/mL. The antioxidant activity of WSAP and APS were better than APS.Exp.two the effects of WSAP and APS on antioxidative of broilersIn order to study the effects of WSAP and APS on the production performance and antioxidative of serum in broilers, the antioxidative was observed in serum treated with WSAP at concentrations of 25, 50, 100, 200, 500, 1000, 2000 mg/Kg respectively and APS at concentrations of 100, 200, 400, 800, 1000, 2000 mg/Kg respectively . The serum were used to investigate the T-AOC, T-SOD, GSH-Px, MDA, hydroxyl radical inhibition and superoxide anion free radical inhibition of lymphocytes by kit. The results showed that WSAP and APS at suitable dosages could significantly promote the production performance, the effects of WSAP is mainly in early growth stage, the effects of APS is mainly in later growth stage. The WSAP and APS at suitable dosages could significantly promote the T-AOC, increase the T-SOD, GSH-Px, inhibit the hydroxyl radical inhibition and superoxide anion free radical, decrease the MDA in peripheral blood and splenic lymphocyte, and WSAP at 500 mg/Kg,APS at 1000 mg/Kg respectively were to best advantage. The WSAP and APS can promote the production performance and antioxidative in broilers. The antioxidative is likely to be one of mechanisms of WSAP and APS to enhance immunity.Exp.three the effects of WSAP and APS on antioxidative of lymphocyte in broilersIn order to study the effects of WSAP and APS on antioxidative of peripheral blood and splenic lymphocyte in broilers, the antioxidative was observed in peripheral blood and splenic lymphocyte treated with WSAP at concentrations of 2, 4, 8, 15, 25, 50,100μg/mL respectively and APS at concentrations of 12.5, 25, 50, 100, 200, 400, 800, 1000, 2000μg/mL respectively. The Supernatant liquid were used to investigate the T-AOC, T-SOD, GSH-Px, MDA, hydroxyl radical inhibition and superoxide anion free radical inhibition of lymphocytes by kit after cultured 24h. The results showed that WSAP and APS at suitable dosages could significantly promote the T-AOC, increase the T-SOD, GSH-Px, inhibit the hydroxyl radical inhibition and superoxide anion free radical, decrease the MDA in peripheral blood and splenic lymphocyte, and WSAP at 15μg·mL-1 ,APS at 100, 200μg/mL respectively were to best advantage. The antioxidative of lymphocyte is likely to be one of mechanisms of WSAP and APS to enhance immunity.
