The objective of this study was to simulating the digestive physiology environment of the intestinal of growing pigs and manufactured a nest of normalization reagents of intestinal digestive zymogram and buffer. validated the effectiveness of the in vitro digestive. Provided basis for quickly simulating intestinal digestive, and also for resolving the key problem of the simulating all gastrointestinal tract digestive for evaluating the nutritive value of the feedstuff.In the first part, we investigated to study the effects of dietary protein content and source on the composition of jejunal fluid in growing pigs, A 4×4 Latin square design was adopted and 4 health and similar weight pigs were feed with four different diets such as corn-soybean meal diet, low protein corn-soybean meal diet, corn-soybean-rapseed-cottonseed meal diet and low protein corn–soybean-rapseed-cottonseed meal diet according to experimental period and pig number in latin sheet. The supernatant of the chyme was collected at low temperature, The digestive enzyme activities and ion concentration of jejunal fluid were determined. The results showed that there were no significant effect of dietary protein source and level on the activity of trypsin, chymotrypsin, lipase , amylase, and on the concentration of Na+, K+, Ca2+, Mg2+, Cl- and pH in the jejunum fluid of growing pigs (P > 0.05). In conclusion, the composition of jejunal fluid from growing pigs can adapt to the dietary CP level and source.In the second part, we study the differences of the method of saturation ammonium sulfate and Directly freeze-dried for reclaiming ratio of enzyme activity. Provide a method for extracting optimal purity and the optimal activity digestive enzyme for dispensing the simulative intestinal fluid. A completely random design was adapted. Six different concentration treatment(30 %,40 %,50 %,60 %,70 %,80 %) for saturation ammonium sulfate method and one freeze-dried concentrated treatment for Directly freeze-dried method. Then Dialysis the salt ions and hybrid-protein by the of auto dialysis apparatus, calculated the reclaim ratio of enzyme activity and total protein .every method made two repetitions, the result showed that Directly freeze-dried method reclaimed the optimal repeatability and optimal reclaiming of the digestive enzyme compared to the saturation ammonium sulfate method.In the third part, we compared the differences of the simulating digestive fluid and the endogenous digestive fluid which from the growing pig jejunal. validate the vitality of the simulating digestive fluid. matched pair design was adapted, use simulating digestive fluid and the endogenous digestive fluid as intestinal period digestive fluid. Determined the digestibility of the dry mater and energy of four feedstuff (corn,Soybean meal,Wheat bran,Cottonseed) and four diets(Soybean meal diet,Wheat bran diet,Cottonseed meal diet,Peanut meal diet) with simulative digestion system. The result showed, the digestibility of the dry mater and energy of the simulating digestive fluid and the endogenous digestive fluid had strong correlation, The absolute deviation between the digestibility was small. This indicated that the simulating digestive fluid had equal digestive ability to the endogenous digestive fluid of growing pigs.
