| In this study, the FAO (FAO) and the International Animal Genetics (ISAG) jointly recommended the 10 pairs of microsatellite DNA primers, the genotype was detected by PCR and gel electrophoresis technology in 4 shaannan Buffalo populations and summed to 192 individuals, and labeled PCR amplified products were analysis by sub-oriented. In order to analysis of the Southern herds of buffalo populations in vivo and genetic variation, the number of alleles and the frequency of specific alleles, heterozygosity, polymorphism information content, the effective number of alleles, and Nei's standard genetic distance (DS), using an average unweighted pair group method and other methods clustering were calculated. The results are as follows:1. Shaannan Buffalo has a rich genetic diversity. 104 alleles were detected in the 10 microsatellite loci, allele frequencies ranged from 0.0208 to 0.4583. The average alleles was 10.4, the alleles number of ILSTS068 loci is the most and reached up to 23, the alleles number of CSSM036 loci is only 6; the rest loci of the alleles number were between 7 and 11. Four groups in all loci are very close to the average number of alleles, which were 9.3, 8.9, 9.1, and 8.5, respectively. In the 10 microsatellite loci, the heterozygosis was from 0.7550 to 0.9288 with an average of 0.8449.2. The unique alleles which were detected were 8, 5, 2, 1 and 0 in 4 Shaannan Buffalo, Chenggu, Nanzheng, Ning Qiang and Han Bin groups. Nanzheng homozygous individuals (frequency 0.0833) in ILSTS068 locus 186bp allele-specific, the remaining alleles were specific to only one individual (frequency 0.0208 or 0.0417) is, that are rare alleles.3. The overall genetic differentiation in Shaannan Buffalo was low, the genetic variation among populations accounted for 1.88% of overall genetic variation. The corresponding total genetic variation within populations was 98.12%, indicating that the total genetic variation among populations of low genetic variation, little genetic differentiation among populations.4. In Shannan Buffalo in the 10 microsatellite loci, polymorphism information content, heterozygosity and effective number of alleles were 0.7007~0.8932, 0.7550~0.9288and 8.7108~13.1469. This study used 10 microsatellite loci are highly polymorphic loci, can be used for the analysis of the genetic diversity of buffalo research.5. Cheng Gu,Nan Zheng,Ning Qiang and Han Bin 4 buffalo populations of Nei's standard genetic distance was 0.0632~0.1168. relatively small, basic at the same level. Decreasing order: Ning Qiang and Han Bin (0.1168), Nanzheng and Han Bin (0.1064), Nanzheng and rather strong (0.1022), and Han Bin Chenggu (0.0884), Chenggu and rather strong (0.0826) and Chenggu and Nanzheng (0.0632).6.In Four groups of Shannan Buffalo, Chenggu and Nanzheng groups together first, then followed with Ning Qiang and Han Bin, clustering its relations with the geographical distribution of buffalo found in Shannan Buffalo patterns and natural environment. |
