In this paper, 11 microsatellite loci were used to analyze genetic diversity and genetic relationship of Guizhou white water buffalo and 6 Guizhou water buffalo. The results showed as follows:1. 72 alleles were tested in 7 water buffalo populations, 49 alleles were shared by all populations and 23 alleles were unshared. The mean number of alleles was 6.5455+1.6348. 69 alleles were tested in Guizhou white buffalo. The allele gene frequencies in each loci were 0.0049~0.5326.2. The percentage of whole polymorphic loci reached 100%. Mean observed heterozygosity, mean expected heterozygosity, mean Nei' s expected heterozygosity and polymorphism information content were 0.9313, 0.7715, 0.7637 and 0.7260±0.0220 respectively in 7 water buffalo populations, and in Guizhou white water buffalo were 0.9414, 0.7822, 0.7784 and 0.7431±0.0496 respectively. There were no differences between Guizhou white water buffalo and Guizhou water buffalo.3. The Nei's standard genetic distance(Ds) and genetic identity were 0.0910~0.3084 and 0.7346~0.9130 calculated by Popgen32 according to the gene frequencies of the 11 loci. Ds between Guizhou white water buffalo and Zunyi water buffalo was the closest(0.0910). Ds between Qiannan water buffalo and Bijie water buffalo were the farthest(0.3084).4. According to stepwise mutation model(SMM) , the time of genetic differentiation of the 7 populations was estimated and analyzed. If the microsatellite average mutation rate was 10"3, the differentiation time between 7 water buffalos was 228~771 years.5. 7 water buffalo populations were divided into 2 groups by UPGMA/Ds. One group included Qiannan water buffalo and Qiandongnan water buffalo, the other includes another 5 water buffalo populations. Guizhou white water buffalo and Zunyi water buffalo were clustered first, then clustered with Tongren water buffalo.6. This study indicated that Guizhou white water buffalo was a mutation population of water buffalo. As to the genetic basis, Guizhou white water buffalo accorded with water buffalo.7. The study provided theoretical evidence to correctly evaluate genetic diversity value of Guizhou water buffalo and constitute feasible conservation strategies of Guizhou water buffalo.
