A Tentative Study On Biological Effect Of Hige-Voltage Electric Of Corona On Scoparium Seed And Mediated Transformation Of Soybean Gy3 Gene Into Scoparium

We have made experiment on the influence of high-voltage electric field of corona in varied doses upon the change of germination rate of scoparrum seed, the content of chlorophyll, as well as the content of SOD and MDA. The result shows that the germination rate of scoparrum seed trends to rise up at first and go down later; the content of chlorophyll, as well as the content of SOD and MDA all undergo an obvious change at certain intensity of the electric field. The index is much higher than their contrasting group.We have also made experiment on the influence of high-voltage electric field of corona upon the water-loss rate and the survival rate of its cell. According to the result, both the water-loss rate and survival rate of callus cell reached an extremely remarkable level when the intensity of electric field comes to 14kv and between 14kv and 15kv respectively.We have also conducted experiment on transferring soybean DNA into the seeds and callus system of scoparium in high-voltage electric field of corona. The result of testing GUS reporting gene shows that when the concentration of this DNA is 200ug/ml, the indicator reached its highest point of 45% after being soaked for 48 hours.The experiment on sensitivity of kanamycin in callus system shows that:the relative growth-rate of callus system is high above 60% when the content of kanamycin is below 30mg/l; the relative growth-rate of callus system tends to be less than 40% when the content of kanamycin is below 50mg/l; when the content of kanamycin is higher than 90mg/l, the growth of callus system are totally restricted.The PCR and RT-PCR examinations on the transformant preliminarily prove that the foreign gene has been transferred into the plant by abstracting fragment from the transformant in the same size with the foreign gene. We also examined the content of crude protein in the transformed seedlings. The result shows:the content of crude protein in the contrasting group is 18.42%, while the quota in the testing group are much higher than the contrasting group with a increase ranging from 2.92%-4.01% and 3.39% in average.
