With the rapid development of my country’s economy,livestock and poultry house breeding is gradually becoming large-scale.At present,most of the livestock and poultry breeding in our country are breeding in houses.Due to the suitable temperature in the livestock and poultry house and the nutrients put in the drinking water,it is easy to form a biofilm on the inner wall of the water pipe.The formation of biofilms can help bacteria resist adverse environments and produce stress-resistant traits.Due to the stubbornness of the biofilm,it is easy to cause a variety of infectious diseases,which is extremely harmful to the health of the animals,and can cause death in serious cases,and cause great economic losses to the farmers.Therefore,the removal of biofilms is the current top priority.At present,biological and chemical methods are generally used to inhibit or eliminate bacterial biofilms.However,the combination of drug residues and other drugs will cause certain harm to animal health.At present,as a new sterilization technology,the high-voltage electric field has the advantages of good sterilization effect,low energy consumption,and no pollution.Therefore,this article aims to study the mechanism of high-voltage electric field on the biofilm,and to explore effective physical methods to inhibit or remove the biofilm.The main contents and conclusions of this research are as follows:(1)Treat the biofilms with different periods and conditions by silver staining,observe and compare the treated biofilms under a microscope,and analyze the mechanism of the high-voltage electrostatic field on the E.coli biofilms.(2)Quantitative study of biofilms by crystal violet staining method.Through SPSS data analysis,it can be obtained that under high-voltage pulsed positive electric field:duty cycle 75%,frequency 200 Hz,voltage10Kv can inhibit E.coli biofilm;duty cycle 25%,frequency 100 Hz,voltage 2Kv can promote E.coli biofilm.Under high-voltage pulsed negative electric field: duty cycle 75%,frequency 200 Hz,voltage-10 Kv can inhibit E.coli biofilm;duty cycle 25%,frequency 300 Hz,voltage-5Kv can promote E.coli biofilm.(3)The growth process of Escherichia coli biofilm is fitted with a cell machine model.The fitting is R2=at room temperature,R2=under high-voltage electrostatic positive electric field,and R2=under high-voltage electrostatic negative electric field.It proves that the growth trend of the E.coli biofilm simulated by the cellular machine model is roughly similar to the growth curve determined by the absorbance OD value.Therefore,it is better to use the cellular machine model to fit the growth prediction of E.coli biofilm.
