With the rising living standards, people pay more attention to their own health. For its smaller side-effects, herbal becomes more popular. Morinda officinalis How, as one of the four Southern medicine, is often used in Chinese medicine. Its main ingredients are anthraquinone compounds, glucides, organic acid, amino acids, oligosaccharides category Iridoids and trace element necessary for the body. It have the effecs of filling kidney, strengthening bones and tendons, alleviating rheumatism and so on. M. officinalis How is an endangered species. With the the development of modern medical technology and the research of M. officinalis How, it has a espanding use and the market demands is large. Because long-time excessive and indiscriminate digging, the wild resources are destructed, the yield and quality of the cultivars can not meet the needs of the market. So the study of good Germplasm's selecte and do some research on its tissue culture not only can provide techniques and methods for the rapid propagation of M. officinalis How, but also can protect the wild resourses. It has a a very important practical significance. The main findings are as follows:1. Fine materials screen. We survey the length of rattan, the number of branches, and stems, the ground, below-ground biomass and the root's polysaccharide content. Then we take the root's weight, the root's polysaccharide content and the root's polysaccharide weight as the main selecte index, measured in 35 plants, from which we selected three excellent material,No.A1, A2 and No. B1 for tissue culture.2. Sterile system sets. This experiment is a pre-test experiment for the experiment of excellent material tissue culture. We studied separately in four areas: the disinfection, drawing month, time and the degree of lignification.(1) Using 10 different sterilization methods on explants steriled. Stat the contamination rate, browning rate and survival rate. The experiment shows that 0.1% mercuric chloride 8min +2% sodium hypochlorite disinfection 10min, their pollution rate is 42.11%, browning rate is 21.05%, which are low, and survival rate o52.63% is the highest. It is the best way to sterile.(2) The month of explant taken has a great impact on the sterile effect, our study found the survival rate in July up to 56.83%, August after, while the survival rate in April and October are less than July and August. (3) The time of explant taken has a great impact on the sterile effect. The results show that in 14:00-15:00 one day which consecutive more than 3 days and no rain to get explants, the contamination rate is the lowest and the survival rate is the highest, reaching 48.89%. It's appropriate to gather the plants.(4) We studied the effect of different degree of lignify and found that not lignified stem explant culture contamination rate is as low as 38.46 percent, the highest browning rate is 37.18 percent; completely lignified stem explant culture contamination rate is up to 64.29 percent and the lowest browning rate was 10.71%; semi-lignified stem explants cultured up to 43.48 percent survival rate and it is suitable for the experiment.3. Tissue culture of the excellent material. In this study, we studied the aseptic system establishment, adventitious bud induction, subculture, seedlings, rooting, hardening and transplanting of the fine materials.(1) Use 0.1% mercuric chloride 10min 8min +2% sodium hypochlorite disinfection method, in order to establish the sterile system of the excellent material. The study shows that the survival rate is 42.1 persent and the result is not bad.(2) In the experiments of adventitious bud induction, when 6-BA concentrations are 0.5mg/L, 1.0mg/L and 2.0mg /L, IBA concentrations are 0.025mg/L, 0.05mg/L and 0.1mg/L, the type MS basic medium of orthogonal design, we found that when 6-BA concentration of 1.0mg/L, IBA concentration of 0.05mg/L, the budding rate is 79.49 percent, which is the highest in all treatments, and the buds grow well. It is the best medium for adventitious bud induction.(3) In the proliferation experiment, take 6-BA , IBA and NAA concentration as three factors, each set up three levels, we found adding 6-BA1.0mg/L+IBA0.1mg/L+NAA0.2mg/L into MS culture medium, the proliferation coefficient is 7.21, which is the highest, and the average seedling height is 3.5cm, the seedlings grow well. It is the the best medium for proliferation of adventitious buds.(4) In the strengthen experiment, take 6-BA , IBA and NAA concentration as three factors, each set up three levels, we find that adding 6-BA0.4mg/L+IBA0mg/L+NAA0mg/L into MS medium, the average seedling length height is 1.988cm, the seedling color is dark green and the stem is strong. It is the best medium to strong seedlings.(5) In the rootage experiment, take the MS medium, IBA and NAA concentration as three factors, each set up three levels, the study found that no rooting hormone 1/2MS medium rootage rate was 100%, and the roots are all normal. It is the best rootage medium.(6)In the M. officinalis How's transplanting matrix choice experiment, after acclimatizated seven days, we found the matrix yellow soil: sand = 1:1, which survival rate is 85.71% , and it is the most suitable matrix for transplant.
