| Acacia melanoxylon is Mimosaceae,the Acacia Mill,woody plants,originating in Australia in the 20th century,the early 1990s the introduction of China,in Guangdong,Hainan planted good performance.Acacia melanoxylon have rapid growth,barren resistance,adaptability,and other characteristics of fine material, available for senior furniture,flooring,decorative material and guesthouses high quality paper pulp,at the same time can use its legume root nodule bacteria symbiotic with the characteristics of fixed into free nitrogen in the atmosphere,continue to increase woodland nitrogen nutrients,some solution to the current forest planted by even the recession caused by the grip,stand a decline in productivity,lower economic issues,Chinese fir,pine,eucalyptus,Schima superba,and other species associated with the ideal of mixed forest.This paper basic materials for Acacia melanoxylon fine clones stem,sterile system from the establishment subculture,rooting induction hardened various links transplant research aimed at exploring Acacia melanoxylon fine clones in vitro culture and rapid propagation of the effective ways to build fast, efficient Acacia melanoxylon tissue culture technology system.Industrial production for providing a theoretical basis and technological support,the findings show that:(1)The suitable explant of tissue culture of Acacia melanoxylon was the top of the new stem with axillary buds.It will differentiate in one week and its pollution rate not more than 20 percent.(2)The pretreated explants of Acacia melanoxylon immersed in 2.5 percent NaC10 for 10~15 minutes or in 0.1 percent HgCl2 for 8 to 9 minutes were sterilized well,contaminated rate less than 20 percent.(3)NAA and 6-BA could promote the differentiation of adventitious buds and showed the highest differentiation rate in the treatment of 6-BA 0.5 mg·L-1+ NAA 0.1 mg·L-1.Its most differentiation rate is 95.3 percent.(4)In the medium pH of 5.2 to 7.0 within the Acacia melanoxylon explants shoot differentiation pilot was found in the pH of 5.8 to 6.4.Acidic conditions more suitable stem bud differentiation,differentiation rate of up to 92 percent while neutral conditions shoot differentiation lower rate,only 38 percent. (5)Of the MS,1/2MS,1/4MS,B5,N6 and improved MS(remove organic compounds)six kinds basic medium of Acacia melanoxylon proliferation and growth of plantlets,MS culture medium was the optimum medium for multiple culture.(6)NAA and 6-BA were the suitable auxins and cytokines respectively in vitro culture of Acacia melanoxylon.They could improve multipication coefficient markedly and the best multiplication efficiency was obtained in the treatment of 6-BA 0.7 mg·L-1+NAA 0.2 mg·L-1.Multiplication coefficient up to 3.26. Light wave could improve multipication coefficient markedly also especially the red wave while light intensity andsugar could not.(7)In vitro Acacia melanoxylon 30d,45d and 60 subculture cycle,the multiplication cycle for about 45 days could both guarantee the quantity and the quality of the adventitious buds.(8)Through the research found that the cooperation of NAA and cane sugar had prominent effects on the adventitious bud height.NAA 0.1 mg·L-1+cane sugar 5%was the best treatment for making the weak buds stronger.(9)Even in the MS medium without any hormones,test tube shoots of Acacia melanoxylon could rooted easily,but the roots showed thin and weak.Adding the auxin in the MS medium could improve the quality of the plant roots.MS+0.1 mg·L-1IBA and 1/4MS+0.5 mg·L-1IBA were suitable for the culture of rooting.The root length and thick degree were moderate and the amount of the roots were large in the two treatments. Light wave could improve multipication coefficient more markedly than light intensity.Red wave is the best light wave.(10)The suitable acclimatization time were 7~9 days.The well-penetrated substance adapted to the plant transplantation in the initial stage and pure vermiculite was the most suitable transplantation substance. |
