| Populus tomentosa×Populus adonpoda, Populus tomentosa×84K Yang (South Korea imported) is a two nice hybrids, obtained by Jun-Feng Fan Researcher., Northwest A & F University, College of Forestry, through crossbreeding experiments from 2002 to 2007.Seedling grew fast in orchard, having strong resistance, but the low survival rate of field cutting, propagation coefficient is not high, limiting the speed of its promotion. Seedling propagation speed can be increased, it's a short-term access to a large number of seedlings. There are many reports of tissue culture about Populus, and more emphasis on a good medium for the screening of clones. There was no systematic report of in vitro regeneration of populus hybrids. The first systematic experimental studies were carried out on the two hybrids, Poplar Populus Tomentosa as female parent of the hybrid .In this study ,taking the stems and leaves of Populus tomentosa×Populus adonpoda and Populus tomentosa×84K Yang as explants, do research on disinfection time, medium, hormone and other tissue factors influencing the course of the study systematically, Initially established efficient regeneration system of Populus×poplars and Populus×84K Young .Thus provides a theoretical basis System for populus hybrids culture and genetic transformation The experimental material collected from the Northwest A & F University, College of Forestry Nursery, overwintering stem ,the water cultured, taking 5-10cm of the spears do explants, experimental results are as follows:1. The best disinfection time for stem explants was: 84 disinfectant disinfection 3.5min, then 0.1% Hgcl2 7min.2. MS medium was the optimal basic tissue culture medium for the new Populus hybrids.3. Stem segment had higher differentiation rate than stem tip, more buds induced than shoot tip.Young leaves had higher induction rate than old leaves.old leaves can only induce d callus, not regenerated shoots.4. The central stem had the best induction effect ,the largest number of induced multiple shoots, and the longest length.5. The best proliferation medium for Populus×adenopoda was MS +6- BA 0.5mg / L + NAA0.01mg/L.The buds induction rate was 93.3%, and the average proliferation number of buds was 6.38 . The best proliferation medium for Populus×84K Young was MS +6- BA1.00 mg / L + NAA0.01mg / L, and the buds induction rate was 90.0%, the average number was 6.00.6. 1/2MS medium was more beneficial than the MS media for rooting, the rooting rate increased as the NAA concentration increased, the over-high NAA concentration is not conducive to rooting. The best rooting medium for Populus×adenopoda was 1/2MS + NAA0.15 mg / L .The rooting rate was 93.3% and the maximum average root length was 7.9cm and the average rooting number per bud was 7.5.The best rooting medium for Populus×84K was 1/2MS + NAA 0.15 mg / L.The rooting rate was 96.7%, and the average root length is 7.6cm and the average rooting number per bud was 7.2.7. Activated carbon controled glass to a certain extent.Medium supplemented with 2.0g / L of activated carbon can make some of vitrification to normal. |
