Populus L. Sect. Leuce Duby and their hybrids have excellent resistance to hard conditions(drought, cold, diseases and pests, etc.) and high landscape value. In northwest region of China, they are main ecological environment construction and landscaping tree species. Due to the close relationship and similarity in morphological characters of varieties(clones) in Populus L. Sect. Leuce Duby, it is very hard to identify them from each other by appearance, which often caused great confusion in production promoting. Doing genetic analysis, building fingerprint and establishing a reliable molecular identification system of the varieties by molecular markers have great significance. 9 varieties(clones) of Populus L. Sect.Leuce Duby(4 new hybrids breeded by Northwest A and F University, their parents and 3relatives) was studied by using SSR and SRAP molecular markers, the results are as follows:1. 106 clear bands were amplified with 9 SSR screened primers, including 89 polymorphism bands,each pair of primers amplify 11.7 bands, the polymorphism rate was83.1%,SSR have high polymorphism; the coefficient in 9 species is between 0.41 to 0.75;According to the genetic similarity coefficient, the Dendrogram of nine species were mapped;Cluster analysis results showed that four new varieties and male parent Populus tomentosa var.runcata first get together, finally together with female parent I- 101.2. 167 clear bands were amplified with 14 screened SRAP primer combinations,including 140 polymorphic bands,each pair of primer amplify 12.84 bands on average;the polymorphism rate was 83.8%, SRAP have high polymorphism;the similarity coefficient of 9species is between 0.4251 ~ 0.7664; According to the genetic similarity coefficient, the Dendrogram of nine species were mapped; Cluster analysis results showed that four newvarieties and male parent Populus tomentosa var. runcata first get together, then get together with female parent I- 101.3. Fingerprint of 9 Poplar varieties(clones) in Populus L. Sect. Leuce Duby was constructed by using selected 3 SSR primers, and 3 SRAP primers with high effective amplification and high repeatability; 9 species(clones) can be indentified by any each one primer alone.4. Compare and analysis of similar coefficient matrix of two molecular markers(SSR and SRAP) and their combination data, it can be concluded that the combination data have high correlation coefficient to each markers, the clustering result is believable.
