The Tissue Culture And Rapid Propagation Techniques On The Stems Of Double Hybrids Of Section Leuce Duby

The study was used tender stems of two Populus alba hybrid as the explants , by means of the proliferation of tufted buds ,to investigate the technology of the induction of axillary bud ,the proliferation of tufted buds, the rooting, the vitrification and other matter of I-101×poplar 84K(Populus alba×(Populus alba×P.glandulosa)) and I-101×Populus davidiana (Populus alba×Populus davidiana) in tissue culture. The chief results are as follows:In the tissue culture of I-101×poplar 84K(Populus alba×(Populus alba×P.glandulosa)) , we can find that MS can be used as the basic medium for induction of axillary buds ,the cytokinin 6-BA has a significant role when the concentration was 0.3mg/L ,the proliferation of tufted buds has a better effects when the concentration of sucrose was 30g/L.The suitable medium for the proliferation of I-101×poplar 84K(Populus alba×( Populus alba×P.glandulosa)) was MS + 0.3mg/L6-BA + 0.3mg/LNAA + sucrose30g/L + agar6g/L. The suitable medium for the rooting was 1/2MS + 0.02 mg/LNAA + sucrose15g/L + agar6g/L.We found the rather higher concentration of macroelement and repeated subculture can cause the vitrification of I-101×poplar 84K(Populus alba×(Populus alba×P.glandulosa)) by the experiment.The experimental study of tissue culture showed that MS can be used as the basic medium of I-101×Populus davidiana (Populus alba×Populus davidiana).The suitable proliferation medium for the tufted buds of I-101×Populus davidiana (Populus alba×Populus davidiana) was MS+ 0.3mg/L6-BA + 0.5mg/LNAA + sucrose25g/L + agar6g/L. The suitable medium for rooting was 1/2MS + 0.02 mg/LNAA + sucrose15g/L + agar6g/L.In China, a nicer foundation for the fast asexual propagation and large-scale industrialized seedling of the two choiceness hybrids (I-101×poplar 84K(Populus alba×(Populus alba×P.glandulosa)) and I-101×Populus davidiana (Populus alba×Populus davidiana)) as mentioned above can be conducted.
