Lily is one of Perennial bulbous flowers, which belong to the Liliaceae of Lilium. It is one of the famous cut flowers of the world.Generally the plants of Liliaceae are diploid (2n= 2x= 24) with two pairs of centric chromosome and 10 pairs of acrocentric chromosome (or telochromosome). China is the center of lily origin and has over halfof all lily species all over the world. It also has a large number of wild species with high genetic diversity. However, a lot of them are in danger of extinction because of natural disasters, pest and so on. While for some reasons, the breeding of lily is undeveloped in China. At present, most of the lily bulbs in commercial use are imported. Currently the varieties of lily become more and more. To study and use the varieties who have good economic characteristics become the focus of lily breeding. Crossbreeding can produce heterosis to raise the growth potential, stress resistance and so on. It also can integrate the good properties of the two parents to make a new variety. A long breeding cycle and the difficulty of interspecific hybridization are the main problems in lily breeding. To select a reasonable group of parents and coordination is the key to the success of hybrid breeding.(1) The best rooting medium was 1/2MS+NAA0.2mg/L and the appreciate sucrose concentration was 30g/L. In this case, the states of and roots were good. The roots are strong and have a great amount. The plants are suitable for transplantation and in situ hybridization.(2) 14 F1-generation strains only have A genome, indicating that their parents are Asiatic hybrids and 6 F1-generation strains only have L genome, indicating that their parents are hybrids of Lilium longiflorum. They all belonged to intraspecific hybridization.None was from Oriental hybrids (O genome).(3) The results of SRAP were that we screened 16 primer compositions and we got 727 DNA bands,76 of which (11.1%) were polymorphic. It means these 20 pro-lily hybrid offspring have different genetic background. Both GISH and SRAP got a same result.(4) This study is to discriminate the Parental genotypes of some pro-lily hybrid offspring whose parental genotype are unknown from the level of chromosome and molecule under the basis of tissue culture. The application of GISH and SRAP at the same time not only can make the conclusions more reliable, but also provide a new way of thinking to determine the parental genotypes of hybrid offspring. Genomic in situ hybridization (GISH) and SRAP molecular markers were used to discriminate the Parental genotypes of 20 pro-lily hybrid offspring whose parental genotype are unknown but can only from A,O,L genome.
