| Lily is an important flower with high ornamental and edible value.It has been extensively cross-bred at home and abroad,and the market demand for ornamental and edible lily obtained through hybrid breeding is increasing.The offspring of hybrid breeding are often bred by rapid propagation technology.The tissue culture balls obtained through tissue culture have the problem of dormancy.Low temperature vernalization is required to break dormancy to normal growth and development until flowering.Some foreign substances can promote The offspring of lily hybrids bloom.Therefore,the research on the propagation and promotion of flowering of ornamental and edible lily varieties is directly related to their industrial production.In this study,the seed balls of the hybrid offspring of Lanzhou lily and Asian lily variety’Trose’and Lanzhou lily and Asian lily variety’Prunotto’were used as experimental materials.The three parts of the study on the effect of exogenous substances on the flowering promotion of the hybrid’Trose’of Lanzhou lily and Asian lily provide the necessary technical support for the industrial production of tissue culture seed balls of the hybrid offspring of lily.Through research,the main conclusions obtained are as follows:1.Tissue culture test results show that for TR(1)the starting medium:MS+6-BA 2.0 mg/L+NAA 0.30 mg/L.(2)the proliferation medium is MS+6-BA 1.0 mg/L+NAA 0.1 mg/L.(3)Rooting medium for strong seedlings:1/2MS+NAA 0.3 mg/L+IBA 0.5 mg/L.(4)Medium for small bulb enlargement:MS+NAA 0.5 mg/L+6-BA 1.0 mg/L+sugar 30g/L+agar powder 6g/L;For PR(1)the starting medium:MS+6-BA 1.0 mg/L+NAA 0.5 mg/L.(2)the proliferation medium:MS+6-BA 0.5 mg/L+NAA 0.5 mg/L.(3)The strong rooting medium:1/2MS+NAA 0.5 mg/L+IBA 1.0 mg/L.(4)Small bulb enlargement medium:MS+NAA 0.5 mg/L+6-BA 0.5 mg/L+sugar 60g/L+agar powder 6g/L.2.The results of the dormancy breaking dormancy test at low temperature show that the hybrid offspring of Lanzhou lily and Asian lily variety’Trose’and Lanzhou lily and Asian lily variety’Prunotto’can be effectively released from dormancy by refrigerating in 4℃refrigerator.TR varieties need 35 day to release dormancy,PR varieties need 42 day to release dormancy.During cold storage,with the extension of cold storage time,the soluble sugar content in the bulb accumulated,the starch content gradually decreased,the endogenous hormone content was actively changed,the phenolic substance content was increased,the enzyme activity of the three phenolic substances changed,the bulb was dormant be broken.3.The experimental results of the flowering promotion study show that:1.6%Ca Cl2solution immersed in the seed ball for 10 hours is the most flower-friendly intervention method for TR.Moreover,different calcium concentrations can improve the flowering quality and flowering period of TR,and have a significant effect on endogenous hormones and carbohydrates in the bulb.Among them,IAA,GA3,soluble sugar and starch have a significant promotion effect on the days from TR to flowering. |
