Four species bulblet of Oriental hybrid lily: Siberia, Sorbonne, Acapulco, Diber, and five species floral organs of Oriental hybrid lily: Siberia, Marco Polo, Acapulco, Casa Blaca', Huangshidai Were selected for in vitro culture. Four stages of primary research were processed, they were: establishment of clone system, multiplication and differentiation, rooting and bulblet formation and transplantation. The results are shown as follows:1. Stage of establishment of clone systemThe results showed that for bulblet, Siberia was the easiest for inducing, which was followed by Diber, Acapulco, Sorbonne and the basal part turned out to be the easiest part for inducing , the second was the middle part, the upper part was hard to induce. Among the inducing mediums of these experiments, MS medium supplemented with 1mg/L BA and 0.1mg/L NAA was suitable for Siberia and Diber; Acapulco and Sorbonne were MS medium supplemented with 2.0mg/L BA and 0.1mg/L NAA.It was found that for floral organ, Siberia was the easiest variety for inducing which was followed by Marco Polo, Huangshidai, Acapulco, Casa Blanca'. The inducing rates from easy to hard of different part in the same variety were: floral receptable, ovary, corolla, filament and style, when the explants of floral organs were cultured in a series of inducing medium. The results showed that MS medium supplemented with 1.0mg/L BA and 0.2mg/L NAA was suitable for inducing Siberia's floral receptable, ovary, style, Marco Polo and Huangshidai's receptable and ovary, Siberia, Marco Polo, Huangshidai's corolla, filament and Acapulco, Casa Blanca's floral receptable, ovary, style, corolla , filament was better in MS medium supplemented with 2.0mg/L BA and 0.2mg/L. When different part of bacteria and germ-free leafs were induced, the results showed that the basal part was the easiest to induce. MS medium supplemented with 3.0mg/L BA and 0.3mg/L NAA were suitable for inducing culture of basal part leaf of Siberia, Marco Polo and Huangshidai.2. Stage of multiplication and differentiationDuring the stage of multiplication and differentiation, for bulblet, it was found that, Siberia and Diber MS medium supplemented with 1mg/L BA and 0.1mg/L NAA was suitable; For Sorbonne and Acapulco was MS medium supplemented with 0.4mg/L BA and 0.05mg/L NAA.For floral organs, Siberia and Marco Polo MS medium supplemented with 0.5mg/L BA and 0.1mg/L NAA was suitable. For Huangshidai, Acapulco and Casa Blanca was MS medium supplemented with 0.3mg/L BA and 0.02mg/L NAA.3. Stage of rooting and bulblet formationDuring the stage of rooting and bulblet formation, the result showed that the category and the concentrations of hormone and sugar affected rooting and forming of bulblets. The NAA Benefited the rooting, the better concentration was 0.5mg/L; The Sugar benefited the bulblet formation, the better concentration was 60g/L; The better rooting and bulblet formation mediun for Siberia, Marco Polo was 1/2MS medium supplemented with 60g/L sugar and 0.5mg/L NAA, Huangshidai, Acapulco, Casa Blanca' and Sorbonne were 1/2MS medium supplemented with 70g/L sugar and 0.3mg/L NAA.4. Stage of transplantationThe stages in transplantation, that was sand bed preparing, domestication, pseudoplantation and transplant soil. The survival rate of transplantation could be 100%.
