Lily is one of perennial bulbous flowers, which belong to the Lilium of Liliaceae. It is one of the famous cut flower in the world. Lily breeding work need long time, hybridization is difficult. In this paper, the oriental lily cultivar "Siberian"(2n=2x=24),OT hybrids "Manissa" (3n=36) and "Robinna"(3n=36)for the material, the parent pollen activity are determined, did conventional hybridization, embryo rescue and hybrids culture, hybrids cytology identification, the results are as follows:1.Pollen viability was determined by the germination method. The results are as follows: oriental lily cultivar "Siberia" has the highest pollen viability, while the pollen viability of OT hybrids was low. The reasons are their complicated genetic background and inconsistent pollen growth. Pollen germination in vitro assay is a preliminary, simple way.In order to improve the success rate of hybridization, it is need to apply TTC method, microscope observe pollen slice and so on.2.Conventional method is used pollination test of different combinations. The results showed that cross hybrids were harvested for the oriental lily cultivar "Siberia" as the female parent, OT lily hybrid "Manissa" as the male parent and "Manissa" as the female parent, "Robinna" as male. To "Manissa" and "Robinna" as the female parent did not receive fruit with embryo.There are two possibilities by inference:First, the style of OT hybrids long, the pollen can not effectively reach the ovary, the conventional method is not advantaged for pollination of OT hybrids. So may apply cut-style-method to overcome pre-pollination obstacles;Second, the selected male parent was poor compatibility to the female parent.3,Screening medium of hybrids and culture conditions, the results show that culture method is better for stripping embryo culture of 60 days mature embryos;suitable medium was MS+GA30.2+ NAA0.1+2% sucrose+0.05% PVP; GA3 is helpful to shorten the germination time, NAA is profit to embryo growth. Proper light is beneficial to early embryo germination.4,We use the methed of cytological identification to authenticate the F1.Blocks for future generations by the possibility of the chromosome reset and exchange,some with parents different forms, the preliminary judge fl hybrid for future generations.the experiment parents.
