In this paper,the ornamental lily varieties and edible Lilium davidii var.unicolor Salisb were hybridized to cultivate new ornamental and edible lily varieties,enrich the economic value and ornamental value of lily,which will provide new ideas for the application of Lily in family horticulture and Lily sea landscape.Firstly,through visual screening,pollen viability identification and stigma receptivity test,suitable hybridization combinations and hybridization time were screened out.Then,the compatibility of different hybrid combinations was evaluated in detail by exploring the pollination methods,the seed setting rate,the continuous observation of seed setting and the fluorescence observation of pollen tube behavior,and the ways of embryo rescue technology and the time of embryo rescue of hybrid offspring were further explored.The suitable mediums were screened out.Suitable hybridization time and mode,embryo rescue time and mode were found,and two hybrid progenies were obtained.The hybrid combinations were Lilium davidi Lilium asiatic ’Tresor’,Lilium davidi×Lilium asiatic ’Prunotto’,and were identified by chromosome number identification and molecular identification.Hybrid progenies were obtained and hybrid lily plants were obtained.Through tissue culture technology,hybrid progenies were propagated and a certain number of hybrid progenies were obtained.At the same time,the soluble sugar and starch content of hybrid bulbs with different refrigeration time were determined,and the change rule of soluble sugar and starch content with refrigeration time was found.They laid a foundation for the follow-up study of new lily varieties.1.Pollen viability test showed that ’Ballroom’,’Prunotto’,’Tresor’,’Rosella’s dream’,’Black Out’could be used as male parents of hybridization.Pollination time of Lilium davidi should be concentrated between 7:00 and 9:00 in the morning.2.When pollinating Lilium davidi×Lilium asiatic Prunotto’,the stigma was treated with 5%sucrose solution first,which could improve the cross compatibility.Pollination at bud stage can improve cross compatibility.The suitable time for embryo rescue was 30 days after pollination.The direct pollination of Lilium davidi×Lilium asiatic Tresor’ was better.Delayed pollination and early pollination were not conducive to improving fruit setting rate.The embryo rescue effect was better at 40 days after pollination,and the stigma of Lilium davidi×Lilium asiatic ’Black Out’ pollen germination solution or 0.1%6-BA treatment was better after natural air-drying for a period of time and then pollination.Delayed pollination and early pollination were not conducive to improving fruit setting rate.The embryo rescue time should be 40 days after pollination.3.The results of pollen tube fluorescence observation showed that there were different degrees of pre-fertilization disorders in all three combinations.4.Ovary section culture efficiency was very low,embryo culture,with embryo sac rescue germination rate was higher,but later survival rate was low,ovule culture method was more suitable for embryo rescue.The ovule culture of Lilium davidi xLilium asiatic ’Prunotto’ needs dark culture first.The optimum medium was 1/2 MS+3%sucrose+0.6%agar+0.01 mg/L NAA+0.1 mg/L 6-BA.The optimum medium for Lilium davidi×Lilium astatic ’Tresor’ and Lilium davidi ×Lilium astatic ’Black Out’ was 1/2 MS+3%sucrose+0.6%agar+0.01 mg/LNAA.5.The sound seedling culture medium for Lilium davidi×Lilium asiatic ’Prunotto’ was MS+lmg/LNAA+2g/L activated carbon+60g sugar+6g agar.The sound seedling culture medium for Lilium davidi xLilium asiatic ’Tresor’ was MS+lmg/LNAA+lg/L activated carbon+60g sugar+6g agar.The suitable hormones for adventitious bud induction in the leaves of Lilium davidi×Lilium asiatic’Prunotto’ progenies were NAA 0.1 mg/L and 6-BA 0.5 mg/L.The suitable hormones for subculture were NAA 0.1 mg/L and 6-BA 1 mg/L.The suitable hormones for adventitious bud induction in leaves of Lilium davidi×Lilium asiatic ’Tresor’ were NAA 0.2 mg/L and 6-BA 1mg/L.The suitable hormones for subculture were NAA 0.1 mg/L and 6-BA 1 mg/L.6.Cold storage for 5 weeks could improve the survival rate of transplanted bulblets.7.Lilium davidi×Lilium asiatic ’Prunotto’ got 15 hybrids,Lilium davidi×Lilium asiatic ’Tresor’ got 12 hybrids.And they were expanded.
