Induction And Identification The Polyploid Seedlings Of Echinacea Purpurea

Echinacea Purpurea is a kind of perennial herbaceous plant of the Compositae,which has good immunoregulation fuction.In this paper the polyploid induction and its identification were studied on the backgroud of the tissue culture results of Echinacea Purpurea. The main results were as follows:1 The best bourgeon conditions were researched.The results showed that the bourgeon effect was the best when the seeds were soaked on the aseptic cotton with aseptic water,and illuminated for 12 hours in 20℃.2 The germinating seeds in advance, the seeds peeping out the radicle and the adventitious buds on callus were induced by the colchicine solution respectively,and the aberrance effects were compared.The results showed that the effect was the best when the adventitious buds on callus were covered with cotton wool infiltrated 0.025% colchicine solution for 24 hours.3 Eleven different culture media were test to induce the mutant to root.The results showed that the culture medium of 1/2MS+NAA0.5 mg·L-1 showed the best rootage effect with the 100% rootage ratio,and thicker and more roots.4 The stomatal apparatus'size, the content of the chloroplast and the activity of the catalase were compared between the normal plant and the mutant also.The statistic evaluation showed that comparing to the normal plant,the stomatal apparatus in mutant were remarkably larger,the content of the chlorophyll was higher in unit of fresh weight,and the activity of catalase was stronger.5 The chromosome number of the aberranced Echinacea Purpurea was determinated.In winter the optimal time of taking the root tip was 9:30am.. Microscopic examination showed that the chromosome of the aberranced Echinacea Purpurea increased evidently,and they were identified as the polyploid.
