Polyploid Induction And Polyploid Identification Of Gypsophila Paniculata

Gypsophila paniculata,one of the most important species of cut flowers in the world.In this study,stem section of gypsophila paniculata of 'ten million starts' was induced by colchicine with three methods.Combined with variety of methods of polyploid identification,was used to evaluation the result of induction.The results were showed as follows:1.The stem section with preculture for 3 days was more compatible for polyploid induction test than CK treatment.Colchicine was used to treat stem section of 'ten million starts' with three treatments in different time and concentration.The results showed that induction efficiency was varied in different treatments.The optimal treatments of soaking method is soaking time with 48 h and 0.05%colchicine concentration.The mortality rate and the induction rate was 73%and 13%,respectively.The mortality was higher with filtration sterilization and high temperature sterilization method.The optimal treatment was the filtration sterilization with 15 d and 0.01%colchicine concentration.The mortality rate and induction rate was 89%and 5%,respectively.2.Microscopic observation showed that:chromosome numbers of contrasts was 2n=2x=34;chromosome numbers of variants was 2n=2x=34.The guard cell length,guard cell wide,stoma length,stoma wide and chloroplast number of variants was significantly higher than CK treatment by measuring stomas with microscopic.3.The stem-tip was more compatible than root-tip in observation with chromosomes of gypsophila paniculata through the observation of cell size and fragmentation index in meristematic zone.Optimized system of observation on chromosomes was soaked by saturated solution of paracide for 4h,fixated by Camoy's fluid I for 24h,dissociated with 1mol/L hydrochloric Acid solution for 15min,and then,stained by carbol fuchsin for 15min,with the stem-tip.
