The effects of low-dose drinking administration of ChIFN-αon immune function of health broilers was studied mainly in this paper.In the experiments 50 young broiles reared to 14-day-old were used,and randomly divided into 5 groups of 10 broilers.During 14~42-day-old the test groupⅡ,Ⅲ,Ⅳwere administrated continuously ChIFN-αin their drinking water followed by 0.1,1.0 and 10U/mL respectively.Ⅰgroup as the control group was only given water.Ⅴgroup as positive control was administrated 50mg/L levamisole hydrochloride.broilers were given drug in their drinking water,then the role of the following was observed:1,The effects of ChIFN-αon cell immunity and the level of Cytokines in broilers after drinking administration.The 15-day-old broilers were given continuously different doses of ChIFN-α(0.1,1,10U/mL)in their drinking water for 21 days.Blood and serum were collected respectively from wing vein in 14d and 21d,the total number of leukocytes was determined by neutral red-crystal violet joint,the differential leukocyte count was measured by the classic method,the lymphocyte transformation capacity was detected by MTT colorimetric analysis,the NK cell activity of killing was measured by LDH release,and the IFN-γ,IL-4,IL-12 level was detected by the double-sandwich ELISA method.The results show that ChIFN-αcan increase the total number of leukocytes in peripheral blood of broiler(sP<0.05),the percentage of lymphocyte(sP<0.01),the percentage of basophils(P<0.01),and reduce the percentage of neutrophils (P<0.01)and monocyte(sP<0.05),the levels of IFN-γ(P<0.05)in serum and the IFN-γ/IL-4 ratio(P<0.05) are significantly improved after drinking administration of ChIFN-α,but the level of IL-4,IL-12 is not significantly affected(P>0.05),the lymphocyte proliferation capacity and the ability of NK cell activity of killing are also not significantly affected(P>0.05).2,The effects of ChIFN-αon immune organs and liver and kidney function in broilers after drinking administration.The 15-day-old broilers were given continuously different doses of ChIFN-α(0.1,1,10U/mL)in their drinking water until 42-day-old.blood was collected from wing vein after weighing,serum was separated for the detection of biochemical index;Then broilers were bled to death for gainning the thymus,bursa,spleen,liver and kidney,Sequently they were fixed by 10% formaldehyde after respectively weighing for the production of paraffin sections.The results are showed as following: (1) 1U/mL group can increase chickens spleen index(P<0.05),and can almost not affect the organizational form of the thymus,the bursa of Fabricius and spleen after drinking administration;( 2) 10U/mL group can increase the concentration of glutamate oxalocetie transaminase(GOT)(P<0.01),total protein(TP)(P<0.01),globulin(GLO)(P<0.01),albumin(ALB)(P<0.01),total bilirubin(TBIL)(P<0.01)and ALB/GLO ratio(P<0.01),1U/mL group is hepatomegaly(P<0.01) , and eosinophil infiltration is caused in mesenchymal liver cells with fatty degeneration of liver cells in 1U/mL,10U/mL group;(3) 10U/mL group can increase the concentration of creatinine(CREA)(P<0.01),1U/mL group can increase the concentration of blood urea nitrogen(BUN)(P<0.05)and lead to renal enlargement(P<0.01),and the varying degree degeneration and nuclear enrichment of the epithelial cell of renal tubular is caused in 1U/mL,10U/mL group.3,The effects of ChIFN-αon the growth and development of broilers after drinking administration.each group was weighed respectively in 14,21,28,35,42 -day-old,then the average daily gain was calculated on the basis of the group.The results show that ChIFN-αcan not significantly affect (P>0.05)the growth and development of broilers after drinking administration.As a result,0.1U/mL,1U/mL group can enhance and regulate the immune function of broilers,and enhance the capacity of resistance to disease in 0~21d after drinking administration of ChIFN-αin broilers.However,the 1U/mL group appears toxicity role in 21~28d after drinking administration of ChIFN-α.10U/mL group can significantly arouse toxic function in broilers,but the growth of broilers is no significantly affected.
