1L-theanine influence to RAW264.7macrophages secretecytokines on miceThis experiment was conducted to investigate the influences of L-theanine toRAW264.7macrophages secrete cytokines on mice.Through the MTT method toevaluate whether the L-theanine have cell toxicity to macrophages RAW264.7, theexperiment was divided into five treatment: a control group,100、200、400and800μML-theanine,each group with5replicates(hole).Through the ecoli lipopolysaccharide(LPS) induced RAW264.7macrophage inflammatory models, use Bio-Plex levitationliquid chip system analysis L-theanine influence to RAW264.7macrophageinflammatory models which have the23kinds of cells secrete factors on mice. Theexperiment is divided into five treatments:a control group,500μM L-theanine,LPSgroup,LPS+250μM L-theanine group, LPS+500μM L-theanine group, each group withthe three replicates (hole).Determination of cytokines,including IL-1α、IL-1β、IL-2、IL-3、IL-4、IL-5、IL-6、IL-9、IL-10、IL-12(P40)、IL-12(P70)、IL-13、IL-17、IFN-γ、TNF-α、Eotaxin、G-CSF、GM-CSF、MCP-1、MIP-1α、MIP-1β、RANTES、KC.Theresults showed that: compared with the control group (0uM, L-Theanine),the L-Theanineof100,200,400and800μM was no significant difference (P>0.05) to RAW264.7; andcompared with LPS stimulation RAW264.7cell model group, L-theanine havesignificant inhibitory effect (P<0.05)on LPS-stimulated RAW264.7cells secreted IL-6,IL-10, IL-12(P40), IL-12(P70), IL-17and TNF-α, the G-CSF, GM-CSF, and Eotaxin.The results indicated that: L-theanine in the range of100~800μM, no toxic effects onRAW264.7cells, and do not affect its growth; L-theanine may adjust the inflammationby inhibit IL-6, IL-10, IL-12(P40), IL-12(P70), IL-17, the TNF-α, G-CSF, GM-CSFand Eotaxin secrete.2The Effects and Mechanism of L-theanine on Growth Performance, Immune Function and ntioxidant properties of BroilersThe purpose of this work was to investigate the L-theanine effects and mechanismon the growth Broilers performance and immune function.690Yellow Chicks about43gwere randomly divided into6groups with5replicates of23birds. The groups weredivided into a control group (non-antibiotic group), antibiotic group,4different levels ofL-theanine (100,200,400,and800mg/kg)groups.(Results)The results showed that:compared with the control group and antibiotic group,the L-Theanine were notsignificantly(P>0.05) effect to Yellow Broiler index of immune organs,antibody levels ofBovine Serum Albumin(BSA),and Newcastle Disease antibody levels;The L-theaninewas improved significantly(P<0.05) to the levels of Secretory Immunoglobulin A (sIgA)of jejunum,serum interleukin-2(IL-2) and interferon-γ (IFN-γ);The activity of serumlysozyme activity had increased after added L-theanine;And no significant effect onserum interleukin-4(IL-4)(P>0.05);L-theanine can significantly increase serum levels oftotal superoxide dismutase (T-SOD) content, and reduce malondialdehyde(MDA)(P﹤0.05); The results suggest that add L-Theanine does not affect the productionperformance and immune organ development of the Broilers,but can significantlyimprove the intestinal sIgA,serum IL-2、IFN-γ and T-SOD contents, reduce MDA.
