The utilization and innovation of bamboo germplasm resources have profound meaning for sustainable development of China's bamboo industry. Phyllostachys heterocycla has the most widely distributed area and high ecological value, so it is particularly important to do the basic research of its germplasm resources. In this paper, taking the mature seeds of Phyllostachys heterocycla as experimental material, the effects of temperature on physiological and biochemical characteristics, the germination property and callus induction of mature seeds was explored. The main results were as follows:1. The effects of temperature on physiological and biochemical characteristics of Phyllostachys heterocycla seeds were investigated during almost a year storage period. The investigating indexes are seed vigor, germination percentage and the relative electrical conductivity。The results showed that the seed germination percentage was found to decline and then rise over the storage duration when stored in different temperature.Overall there is no obvious difference in seed germination percentage. So the result is conferred that Phyllostachys heterocycla seeds go into secondary dormancy during the storage. There was a negative correlation between the seed vigor and the relative electrical conductivity, a positive correlation between the seed vigor and germination percentage. The relative electrical conductivity and germination percentage are negative correlation. Especially under freezing condition, the correlations between different indexes are relatively strong. The difference of the relative electrical conductivity stored in different temperature happened earlier, it maybe the sensitive parameter in assessing seed quality. After almost 1 year of storage, the different temperature has little influence on seed vigor and germination percentage.2. The germination property, the effects of endosperm and sucrose on seed germination of Phyllostachys heterocycla was studied. The results indicated that, The accompany fungus of the bamboo seed endosperm is the main pollution factors in the operation of tissue culture, the less the amount of endosperm carried on the seed is,the easier the disinfestations controlled; The amount of seed endosperm does not affect seed germination in the culture medium and by increasing the sucrose density also does not change its germination rate obviously; 3. There had no relation between callus inductions with germination of seed in the process of callus induction. Adding different plant hormones have effect on the callus induction frequency and the callus quality. The callus initiation and quality may influenced by the mutual impact and function of the induction medium , plant hormones and many other kinds of factors.4. Isolated main fungal reducing strains from the seeds of Phyllostachys heterocycla, 8 strains were isolated. Analyzed their ITS sequences, three strains of them were identified.
