9 Min, 48 Beijing Black, 48 Landrace, 48 Large White and 48 Laiwu pigs were selected in this experiment. AS-PCR and PCR-RFLP technologies were usd to detect mutation loci and investigate the polymorphism of H-FABP promoter in Min, Beijing Black, Landrace and Large White pigs. And also, the the polymorphism of MC4R D298N mutation loci were investigate in Beijing Black, Laiwu and Large White pigs. The results indicated:There is a new SNP (T>C) at -224bp, named H-FABP-proT224C, from the initiation codon (ATG) in H-FABP promoter region in Min, Beijing Black, Landrace and Large White pigs. And in min pigs group, the genotype frequency of TT, TC and CC were 0.22, 0.67 and 0.11 respectively, the gene frequency of T and C are 0.56 and 0.44 respectively which follow Hardy-Weinberg equilibrium. But in other groups, only CC genotype appared. And as the SNP was found only in Min pig which has more intramuscular fat content (IMF) provide the support that H-FABP-proT224C could play an important role on IMF.To MC4R D298N, in Beijing Black and Large White pigs group, the gene frequency of allele D and N are 0.51 and 0.49, 0.57 and 0.43 respectively, which can also follow Hardy-Weinberg equilibrium, while none polymorphism has been investigated in Laiwu pigs group. We can infer that just because the D298N mutation in MC4R fragments brought low IMF and high Lean Rate to introduced breeds like Large White pigs and developed breeds like Beijing Black pigs. D298N mutation in MC4R maybe also plays an important role on IMF.
