DNA samples from 265 pigs including four Chinese local breeds and three imported breeds and one cultivated breed and wild pig were analyzed on their genetic variation and genetic relation using modem molecular markers technology. The study has two aims. The one is that relationships of eight pig breeds and wild pig was studied by RAPD molecular marker, the other is that the genetic variation in pig H-FABP gene 5'-upstream region and the second intron region were detected by PCR-RFLP. The future attempt of our study is to look for a possible relationship between different genotypes in H-FABP gene and variation in IMF content, further to investigate whether H-FABP locals is the major gene for IMF deposition.20 arbitrary primers which contain 10bp per primer were screened by amplified polymorphism DNA (RAPD). The results showed that 14 Primers amplified reproducible DNA fragments, 8 out of 14 primers can amplified signer fragments, 6 out of 14 primers can amplified signal fragment. The number of amplified fragments to primer ranged from 2 to 11, 4.1 polymorphic fragments per primer. The study found 14 markers using the 4 primer and investigated the genetic relationship of eight pig breeds and wild pig. The results were as follows.1. This study showed that the genetic distance derived from Nei's method in 3 imported breeds were the closest, respectively 0.0200,0.0426 and 0.0638, BaMei pig, HanJian pig., NeiJiang pig genetic distances were closer than that of RongChang pig and HanBei pig, the genetic distance is the farest between Landrace and wild pig.2. The cluster trees based on the mean genetic distance from Nei's cluster trees method software were constructed by NJ methods. The result showed that three imported breeds ware clustered first and their relationship were closest; in three Chinese breeds, Bamei and Hanjiang pig which their relation ship was closer were clustered, then cluster with Neijiang pig; Rong chang and HanBei pig was a cluster, then cluster with Chinese breeds; nowever, wild pig was clustered with Chinese bread firstly, then clustered with imported breeds.Genetic variations of 5'-upstream region and the second imtron in 265 pigsH-FABP gene were detected by PCR-RFCP molecular marker technology. The result showed:1. Hinfl-RFLP in 5'-Upstream(1) Hinfl-RFlP in 5'-upstream region was verified in 8 pig breeds and wild pig, and the frequencies of H allele in DU, CH, DA, NE, BA, HH, RO, HB and YZ were 0.7500, 0.7188, 0.9167, 0.3333, 0.1250, 0.6909, 0.1167, 0.8500 and 0.9375. The genotypes distributes in DA, HBandYZ,DU, CH, and RO, BA and HH were similar (P>0.05), and that of different types were striking (P<0.01).(2) A chi-square analysis suggested that, except for HH and YZ (PO.01) for Hinfl-RFLP allele frequencies and genotype frequencies of the rest breeds population were in Hardy-weinberg equilibrium (P>0.05).(3) For Hinfl-RFLP, DA, BA, HH, HB and YZ had low polymorphism (PICO.25), the other had intermediate polymorphism (0.25
