The experiment was conducted with the softwood of red willow,malva sylvestris L and sorbus aucuparia, chrysanthemum, sweet potato test-tube seedling. After cutting some days, the survival rate, rooting percent and number of root were studied during rooting of five kinds of plant's cuttings by treating of different growth regulators with water control. And sorbus aucuparia, chrysanthemum, sweet potato tissue culture seedlings were used as reseach material for out test-tube cultivation.From this experiment, we could draw the conclusion as follows. Firstly,the MS solution and 10-3Junshabaoguo adding in rooting regulator could improve rooting of cuttings.And the effects of different rooting regulator on five kinds of plants were different.Red willow and malva sylvestris L were obtained the best rooting rates, length of root and the number of root with NAA 100mg/L+ IBA100mg/L+6-BA10mg/L +B610mg/L+1/2MS +1000-1 Junshabaoguo . The rooting rate,length of root and the number of root of Red willow with this treatment were increased by 7.1%,57.1% and 72.5% separately.Sorbus aucuparia, chrysanthemum test-tube seedlings were obtained the best rooting rates, length of root and the number of root in 1000mg/L IBA.Secondly, the results showed that MP could improve rootting and growth of the five kinds of plants.Five kinds of plant cutting were obtained the best rooting rates, length of root and the number of root by the use of growth regulars and MP(1/2MS+ MPC20mg/L+CVB200mg/L). The survival rate and rooting rate of sorbus aucuparia with this treatment were increased by 199.99% and 9.3% separately. Thirdly,the physiological biochemistry metabolize changes were studied durring rooting of sorbus aucuparia and chrysanthemum test-tube seedling which were treated with growth regulator and MP.The result showed that the contents of soluble carbohydrate, soluble protein and chlorophyll were all increased gradually with the treated days, and the activities of SOD were also enhanced.This showed that MP increased survival rate and rooting rate by regulating the metabolism of cutting's physiology and biochemistry.Last,it was found that the quality of test-tube seedlings,,culture media and growth hormones had a remarkable influences on out test-tube rooting of the tissue culture seedlings.Compared with test-tube rooting,out test-tube rooting could improve the rooting quality and the absorption, improve the resistance,the transplant survival rate .
