Virus disease is one of the major diseases that affect livestock development at present.many researehes have showed that many the active ingredient of natural plant have good antiviral activity; Ammopiptanthus is a kind of natural plant.It has been reported showed that the alkuloids of Ammopiptanthus have antiviral activity, in order to further research the antiviral activity of Ammopiptanthus alkuloids and security, the experiment in vivo and in vitro antiviral and subacute toxicity Been studied separately.The obtained main contents are as folloes:1. Effect of total Alkaloids of seeds of Ammopiptanthus BPIV-3in vitroThe cytotoxicity of total alkaloids of seeds of Ammopiptanthus mongolica to MDBK was assayed by observing cytopathic effect and MTT colorimetry, RBV was used as positive reference drug,Abstract:(Objective):though four administration routes of virus and drug, cpe of MDBK cells induced by BPIV-3was observed and OD value was assayed according to MTT,meanwhile,TC50、IC5、TI Be calculated.(Result) The results showed that The drug safety concentration is0.31mg/ml,TC50is2.19mg/mL,BPIV-3TCID50is10-7.733/0.1mL,IC50is0.25mg/mL,the maximum inhibition ratio is66.15%,TI is8.76,about the mechanism of direct action;whileIC50is0.203mg/mL,the maximum inhibition ratio is52.91.%,TI is10.79about the mechanism of inhibition on virus proliferation; meanwhile,IC50is0.16mg/ml,the maximum inhibition ratio is63.38%,TI is13.69about the mechanism of prevention effect on virus adsorption; while,IC50is0.085mg/ml,the maximum inhibition ratio is86.49%,TI is25.76about the mechanism of comprehensive inhibition effect directly. These results indicated that the total alkaloids of seeds of Ammopiptanthus mongolicus has an obvious effect against BPIV-3virus in vitro.2. Effect of total alkaloids of seeds of Ammopiptanthus on BPIV-3in vivoIntranasal BPIV-3virus infection in mice to establish animal models of virus infection, tested on effect of total alkaloids of seeds of Ammopiptanthus mongolica on BPIV-3in vivo,mice were divided into four group at random as administering group, normal control group,model control group,and positive reference group(RBV),some parameters were measured,for example death rate,average life days, lung indx and the inhibition ratio to pathological changes in lung、liver, spleen, kidney.the result showed that the survival time in the administering group of mice was markedly prolonged and the rate of Protection against death is50%at the concentration of14.72mg/kg, lung indx and the inhibition ratio, the survival time is significant difference compared to the control group fed with ribavirin.the finding suggest that the total alkaloids of seeds of Ammopiptanthus mongolica has a strong anti-viral effect.3. Study on Subacute Toxicity of total alkaloids of Ammopiptanthus in micesAccording to the Acute Toxicity result,the experiment divides into three dose groups and one control groups, total alkaloid gives to mices in each dose everyday with a metal tube for4weeks.The mices in control group eat food and drink water freely, the behavior such as drinking water, appetiting, major organs and morphological structure were observed.The weight changes and physiological and biochemical parameters were measured.Results:there were no abnormal changes on spirit activity, eating food, drinking water, compared with those in control groups, histological studies showed that the treatments did not induce any pathological changes in the heart, liver, spleen, kidney and lung of the animals. Conclusion:the total alkaloids of Ammopiptanthusmongolicais belongs to low-toxic drugs and the safety dose is14.72mg/kg.Key words:the total alkaloids of Ammopiptanthus; subacute toxicity; Bovine...
