The chemical compositions of the fruiting bodies of Ganoderma applanation(Pers.)Pat were extracted systemically by solvent extraction,solvent extraction;Simultaneously protective effect of components isolated from G.applanatum as subjects against carbon tetrachloride(CCl4)-induced liver injuries,their anti-inflammatory and analgesic effects were studied in mice;The petroleum ether component and the ETOAC component of the fruiting bodies of G.applanatum were isolated systemically by TLC,silica-gel columu chromatography and Sephadex LH-20 columu chromatography.First,solvent pickling process was adopted to extract organic compounds from the fruiting bodies of G. applanatum.The dry fruiting bodies of G.applanatum was smashed into powder.The powder was extracted with petroleum ether for 3 times in stainless steel drum at room temperature,the extracting solution was collected and combined,then distilled under reduced pressure at 40℃.The petroleum ether component of G. applanatum was obtained.The residual powder,in which Pertolemu ether has been evaporated,was extracted 3 times with 95%EtOH.The ditto mehotd was employed in this poreess.EtOH component of G.applanatum was obtained.The EtOH extract was dissolved in water,then the suspension was extracted with ethyl acetate and n-butanol in turn,the ETOAC component and the n-butanol component of G.applanatum were obtained respectively.The polysaccharides was extracted from the fruiting bodies of G.applanatum by the traditional method, Content of polysaccharides was detected by phenol-sulphuric acid method,the result indicated that polysaccharide content of the extract was 57.1%.The acidic component of G.applanatum was extracted by solvent extraction.Next,using a model of carbon tetrachloride-induced liver injuries to study initially liver protective effect of components isolated from G.applanatum,including the petroleum ether component,the ETOAC component,the n-butanol component,the polysaccharides and the acidic component.The result indicated that the petroleum ether component at a dose of 0.1 g/kg,ETOAC component at a dose of 0.2 g/kg and the polysaccharides at a dose of 0.4 g/kg could significantly decrease the activities of ALT and AST in blood serum(P<0.05,P<0.01).It showed that they had obviously protective effects on liver injury induced by CCU in mice.The activities of ALT and AST had significantly enhanced following dose of the petroleum ether component increased(P<0.05).It explained that the petroleum ether component had the negative action on liver.The effects of the acidic component and the n-butanol component were similar that they did not show protective effect on liver.Appling to ear swelling with Xylene,acetic acid writhing test and hot plate test in mice to study the anti-inflammatory and analgesic effects of 5 components isolated from G.applanatum.The result indicated that the petroleum ether component,the n-butanol component and the polysaccharides can inhibit swelling level of mouse auriele induced by xylene in mice(P<0.05).the n-butanol component has the most strongest anti-inflammatory effect in them,of which swelling inhibition ratio of mouse auriele induced by xylene was 30.2%.the polysaccharides has expressly analgesic effect induced by Chemical Stimulation(0.6%acetic acid),can inhibit writhing response frequency induced by acetic acid,of which analgesia ratio was 37.2%.the polysaccharides need more longger Metabolism time to develop analgesic effect with physical Stimulation(hot plate test),can significantly improve the pain threshold of mice,meanwhile analgesic effect of the polysaccharides was more stronger than aspirin.The effects of the acidic component and the ETOAC component were similar that they did not show anti-inflammatory and analgesic effects.Last,the petroleum ether component and the ETOAC component of the fruiting body of G.applanatum were isolated systemically by TLC,silica-gel columu chromatography and Sephadex LH-20 columu chromatography.The petroleum ether component was subjected on silica-gel with a petroleum ether-ETOAC system elution,compound 2 was obtained,through sephadex LH-20 column chromatography with a ETOAC elution,compound 1 was obtained.The ETOAC component was subjected on silica-gel with a CHCL3-ETOAC system elution,through sephadex LH-20 column chromatography with a MEOH elution, compound 3 was obtained.Compound 1-2 were identified as Unsaturated Fatty Alcohol,ergosterol through TLC,NMR spectrum and MS,et al.Compound 3 needed to be identified further.
