Rosa davurica Pall.grows in China's Greater Hinggan Mountains,Smaller Hinggan Mountains Changbai Mountain,is a kind of abundant resource,and has the good value for developing and utilizing.But current research reports on Rosa davurica Pall.mainly in the composition of its fruit and processed products,such as soft drinks,tea,cakes and candy and so on,it is very lack of the research and utilization of its stems and leaves part.And the stem of Rosa davurica Pall.is rich in flavonoids which have antioxidant,anti-aging,treatment of cardiovascular and cerebrovascular diseases,anti-bacterial anti-inflammatory and other physiological functions.In this paper,we chose Rosa davurica Pall.stem as raw material, studied on extraction,separation and purification of flavonoids aglycones in Rosa davurica Pall.as well as antioxidant in vitro.The main results in this paper were as follows:1.The preliminary experiment of chemical composition in Rosa davurica Pall.stem.Plant chemical composition system preliminary experiment method was used to identify that the Rosa davurica Pall.stem contains polysaccharides and glycosides,phenols,flavonoids, coumarin and within lipid,tannin,organic acid,saponins;it also may involved amino acids, peptides,protein,anthraquinones compounds;it did not comprise alkaloids,cardiac glycosides, steroids and triterpenes.Special preliminary experimen determined the flavonoids in Rosa davurica Pall.stem was flavanone(alcohol) type.2.The studied on extraction technology of agiycone flavonoids in Rosa davurica Pall. stem.First of all,the hydrolysis conditions of flavonoid aglycone was inspected and its best result was that the ratio of hydrochloric acid to the raw materials was 4:1,concentration of hydrochloric acid was 3mol/L,the hydrolysis time is 2h,the hydrolysis temperature was 90℃.Following,selected two types of solvent to extract,and then hydrolyzed for aglycone flavonoids.After trial we indicated that based on the single-factor test we designed the experiment with the method of orthogonal test and found the best conditions of extraction using ethanol as solvent were that raw material particle size was 100 to 120 head, concentration of ethanol solution 60%,extraction temperature was 80℃,solid-liquid ratio was 1:15,extraction time was 3h.Under these conditions the extraction of flavonoid aglycone rate was 93.36%.Through single-factor test we determined the sodium acetate solution as the best extraction solvent extraction conditions were as follows:raw material particle size was 100 to 120 head,the concentration of sodium acetate aqueous solution was 6%,extraction temperature was 90℃,solid-liquid ratio was 1:20,extraction timing was 4h.Under these conditions the extraction of flavonoid aglycone rate was 85.49%.Compared the extraction conditions and results of two solutions we could be seen:two types of solvent extraction conditions was similar,but rate of extraction was different.So we chosed ethanol solution as extraction solvent.3.The studied on flavonoid aglycone purification Rosa davurica Pall.stem.We chose from eight kinds of macroporous resin and through the comparison of static adsorption to screen the suitable kind for the flavonoid aglycone purification Rosa davurica Pall.stem,and studied on the static adsorption,desorption characteristics of this resin and determined the dynamic adsorption and desorption process.The results show that the D4020 resin has a higher adsorption and desorption rate than others,so it was suitable for purifying flavonoid aglycone in Rosa davurica Pall.stem.The best conditions of adsorption and desorption were as follows: stock solution concentration was 0.9mg/mL,pH value was 4.5,sample flow rate was 1mL/min, ratio of diameter to height was 1:15;eluate was ethanol solution of 70%,pH value was 9.0 and with 50mL of eluate have basically been able to elute down the flavonoid aglycone in Rosa davurica Pall.stem.The purity of flavonoid aglycone became 57.3%after purified compared to 22.4%before.It said that D4020 macroporous resin played a better effect on separation and purification of Rosa davurica Pall.flavonoid aglycone stems.4.The studied on antioxidant activity of aglycone flavonoids in Rosa davurica Pall.stem. We respectively determined the scavenging capacity of total antioxidant,hydroxyl radical, DPPH·free radical removal and anti-lipid peroxidation ability of aglycone flavonoid extraction in Rosa davurica Pall.stem.The results showed that flavonoid aglycone in Rosa davurica Pall. stem has a good antioxidant activity,and flavonoid aglycone through resin purified was better than raw flavonoid aglycones in antioxidant activity.
