Rosa roxburghii Tratt. is a kind of rosaceae rose deciduous shrubs that throughouts on mountainous and hilly terrain ranging from 1000 to 1600 meters elevation. Which is also a kind of wild resources in yunnan-guizhou plateau has strong adaptability, easy to grow, and contains a lot of flavonoids, vitamins, polysaccharide and other pharmacological compositions. But the utilization of rosa roxburghii tratt. is limited to preserved fruit, fruit juice processing now, and relevant research reports lessed systemic and depth. The article based on rosa roxburghii tratt. collected from guizhou as raw material, studying on extraction, separation and antioxidant activity of flavonoids from rosa roxburghii tratt.(FRR), and aimed at providing a theoretical basis for the comprehensive exploitation and utilization.The main research contents and conclusions were listed as followed:(1) With the yield of FRR as indicator, studying effects that ethanol concentration, solid-liquid ratio, extraction temperature and extraction time by single method tests and response surface methodology (RSM). The result showed optimal conditions for SBE were 80% ethanol,1:15 (g:mL) of solid-to-liquid ratio, and extracting at 90℃ for 80:40:40 (min), the extraction yield of flavonoids under these conditions was 27.70 mg/g, which was equivalent to the mass fraction 8.41%.(2) Investigating the adsorption characteristics of FRR by eight kinds of macroporous resin, the result showed AB-8 macroporous resin was the best resin used in refining FRR. Through static test and dynamic test, determine the best craft: post specification 1.6 by 50 cm, and diameter to height ratio 1:22.5. Adsorbate solvent:water, pH was natural; Eluent:80% ethanol, pH of 6; Sample concentration: 5 mg/mL; sample speed:1.5 mL/min; sample amount:8.5 BV; elution rate:1.5 mL/min; elution volume:2.5 BV. The contents of FRR reached more than 37.24% after refining, and increased 4.4 times.(3) Using high performance liquid chromatography (HPLC) method to seperate FRR further, got Ⅰ, Ⅱ components. By UV spectra, HPLC and mass spectrometry were preliminary qualitative analysis, and the results show that the two components were divided into flavonoids, and component I for quercetin, component II for resurrectionlily phenol.(4) Investigating the activities of crude FRR, refined FRR, and two components of FRR, and the activities were linear with the concentration of 4 samples increasing in range significantly. Results show that reducing power, total antioxidant capacity and scavenging ability of free radicals (· OH, ·O2-) of crude extraction was lowest among refining FRR, component Ⅰ and Ⅱ; DPPH-radical scavenging ability of crude FRR was higher than refined FRR and two components, it may be related to other active ingredients it contained; Crude FRR, refined FRR and two components all have ability to resist lipid peroxidation, and the difference was not significant; Inhibitory rate (IC50) results showed that, the concentration of FRR and antioxidant activity have linear relationships in some range, and the IC50 value of the sample and rutin is approximation; The less the IC50 value, the antioxidant activity lower, so the result indicates that FRR has strong antioxidant activity.
