Screening Out Rumen Anaerobic Fungi Of Efficient Cellular Enzyme And The Study On Utlization Of Fiber Material

For many years, it was focused that ruminant could degrade crude fibre effectively. Degradation of fibre owed to rumen microorganisms. In fact rumen anaerobic fungi plays a very important role on the degradation of crude fibre. As a result, it was necessary to study rumen anaerobic fungi for improving availability of ruminant to crude fibre. In this study, 8 strains anaerobic fungi were separated, purified and identified from rumen, and further were studied their fibre digestibility and apposite preservation condation. The results showed that:1. 8 strains rumen anaerobic fungi were separated and purified from rumen of Inner Mongolia cashmere goats, Guanzhong milky goats, Holstein cows and Mongolia sheep in this experiment by Hungate roll tube method. Through identification A1,B1,C1,D1 strains rumen anaerobic fungi were Neocallimastix, while A2,B2,C2,D2 strains rumen anaerobic fungi were Piromyces.2. A1 and D2 had strong fibre digestibility and cellular enzyme activity. When the both were cultured using cellulose and cellobiose and xylan as substrates carbon source, respectively, the CMC enzyme activity was rather highest using cellulose as substrates carbon source than using cellobiose as substrate carbon source.3. A1 and D2 were cultured using five different natural roughages as substrates carbon source. The results showed that the interactions between gas production or CMCase activity and the degradation rate of fibre were not always linear correlation. Piromyces strains and Neocallimastix strains have similiar fiber degradation activity.4.By preservation experiments, we found the preservation method was better using 12% glycerol at -20℃in liquid, but the enzyme avtivity and live fungi counting were both unstable, at the save time, preserved time was no more than 90d, so the results of preservation experiments was far from perfect.
