, Abstract: Isolation and purification of rumen anaerobic fungi which produced cellulase were presentend in the paper. Screening of high activity cellulase of one strain WAl from rumen anaerobic fungi was identited.The enzyme property and mutation and the effects of fermentation transform to fibre of wheat bran were studied in the paper.The results were as follous.1 .Selection of rumen anaerobic fungi with cellulase activity was the purpose. The methodology of fluid medium cultivated for promote fungi, and CMC-Na agar anaerobic cultivate for screen strain were adopted. Then Congo—Red stained and the ascertain on relative ratio of cellulase activity with H:C value(ratio of diameter of clearing zones to colony) were applied. The results showed that, one strain of anaerobic fungi with high cellulase activity was selected. It was identified as Orpinlmyces. The optimum pH of FPase and CMCase and salicinase respectively were 5.0.5.5 and 5.0.The stability pH range of FPase and CMCase and salicinase were respectively 4.0~7.0,4.0~7.5 and 4.5~7.5. The optimum temperature were 55℃,50℃ and 55℃. The best thermal stability of the enzyme was salicinase, then CMCase, and FPase was a curve like S as synmaifest of three cellulase and it's cellulase activity was 117U/ml on the optimun condition.2.Selection of rumen anaerobic fungi with high-yield cellulase after mutagenesis was the purpose. The rumen anaerobic fungi spore was treated by UV,NTG and UV+NTG in sequence. Then the mutated strains were first selected by H:C. And the fatality rates were 68% and 74% respectively when the anaerobic fungi spore were treated by UV 100s and NTG 30min. This was the suitable dose which rumen anaerobic fungi spore by complication mutation. The results showed that 10 strain of rumen anaerobic fungi with relatively high H:C value were obtained after solid medium cultivated. And after lOtimes...
