OBJECTIVEL.Sabrazesi belonged to Plasmodiidae,Leukocytozoon.Fowl blood cells and fowl internal organ cells were parasitized.The harm is larger to egged fowls and growing fowl,it could effect the growing and egged function;It had also many disservices to chicken.Chicken would dead,when attack rate was high.Enormous economy loss was there to poultry industry.Meanwhile,the study of Leukocytozoon majored in L.caulleri.The study of L.Sabrazesi was litter.So this study was to aim at consummating epidemiology,aetiology,blood biochemics of L.Sabrazesi in Hunan, constructing the different animal model of L.Sabrazesi.METHODS1.In dyeing of L.asbrazesi,the 12th method(Gram staining,Methylene blue staining, Acid fast staining,Wright staining,Giemsa staining,Azalein staining,Azalein staining,Wright-Giemsa staining,Rickettsia staining,May-Grunwald staining, May-Grunwald-Giemse staining,Eosin-MB staining) were used and compared.2.In epidemiologic survey,the infection of L.asbrazesi on aniso-age years fowls,aniso-vatiety fowls,aniso-breeding ways were deeply invested with blood smear;the information of buffalo gnat was initial invested with prehension.3.In different animal model,the infected fowl blood of L.asbrazesi was vaccinated with different ways to chicken and duck.The period of appearance,peak,disappearance of L.asbrazesi was observed.Numbers of gametophyte on the period of peak,proportion of female gametophyte and male gametophyte on the period of peak,the diurnal variation of gametophyte were also viewed.4.In biochemical indicator,the body weight of chicken and duck were measured.The periphery hematology was determined.When the animal were infected on 70th,the serum biochemical indicator was determined,the organ weight was surveyed,clinical pathology of infected animal wad taken notes. RESULTS1.The results indicated that Wright-Giemsa's,May-Grunwald-Giemse's,Eosin-MB's were the best methods and Gram's,Methylene blue's,Wright's,Giemsa's, May-Grunwald's,Rickettsia's were the better,while Acid-fast's,Azalein's,Albert's were the worst.The result of Methylene blue's was better by the short staining time, handling simply,conspicuous layer,the most integrated structure,the more limpid structure and the high detection rate.Wright-Giemsa's,May-Grunwald- Giemse's, Eosin-MB's were the double stainings.The result were best by conspicuous layer,the most integrated structure,the most limpid structure and the long staining time.2.The sum of survey was 365,infection rate was 26.5%(0.5%-85.7%),average leuk-infection rate was 3.0%-10.3%.The area of highest infection rate was Jishou.The infection rate of Changsha fowl(37.5%) was higher than local fowl(31.3%);The infection rate of meta-fowl was the highest(42.9%);The infection rate of free-breeding fowl(27.9%) was higher than confined-fowl(22.5%);The size of male gametophyte was 11.0um×17.4um,the size of female gametophyte was 11.6um×18.3um.The sum of Simulium metatarsale was 17,infection rate was 23.5%.3.L.Sabrazes appeared at post infectionem 36th on the animal model of chicken.This illustrate the animal model of chicken succeed.The infected duck did not detect L.Sabrazes on 70th.This illustrate the animal model of duck unsuceed.The period of appearance was 36th,the period of peak was 49th,the period of disappearance was 60th.On the peak time,the ratio of female gametocyte and male gametocyte was 86:65.Meanwhile,the gametocyte in day was more than night.The acidophilic cell(added 6 double) and basophilic cell(added 2 double)of infected poultry added.4.The growing of body was not conspicuity discrepancy with control group(P>0.05).This shown un- influence to chicken and duck at the experimental condition.TP,ALB were not conspicuity discrepancy with control group (P>0.05),ALT,AST,ALP were also not conspicuity discrepancy with control group.This shown un- influence to chicken and duck at the experimental condition.When the animals were infected on 70th,thoracic gland and spleen and bursa of Fabricius were not conspicuity discrepancy with control group(P>0.05).CONCLUSIONS1.Wright-Giemsa's,May-Grunwald- Giemse's,Eosin-MB's were the best methods.Methylene blue's was fit for the epidemiology investigation;Eosin-MB's was suitable for application in countryside in stead of Wright's,Giemsa's and Wright-Giemsa's. 2.The epidemic of L.Sabrazes was all over Xiangxi,the 4 of 5 towns have the distribution of L.Sabrazes.The infected rate were from 0.5%to 85.7%.As the location around the dam,where suited the buffalo gnat growth,the infected of the village of Tang Ba was highest(85.7%).Meanwhile,the investigation shown that Changsha fowl and middle fowl was easiest infected.This was different from the conclusion of Lin Yuguang which shown chicken was easiest infected.This would was concerned with investigate place and season.The literature of the infected rate of free-fowl was concorded with the investigation which shown the infected rate of free-fowl was higher than confined-fowl.The result of gametocyte was agreed with Liu Yi.The collection information of buffalo gnat illustrated it was the spread intermediary of L.Sabrazes.3.The animal model of chicken was modeling to succeed.The period of early appearance was 36th,the period of late disappearance was 70th,the proportion of female and male gametocyte was 86:65.These were differet with the literature and data which shown the period of appearance was 8.5th,the proportion of female and male gametocyte was 260:240.The different infected ways and different infected fowl species would lead to.The diurnal variation of gametophyte was the periodism of the day high,the night low.This was according with Noblet'report of L.simondi.4.The biochemical indicators were not conspicuity discrepancy with control group.This shown un- influence to chicken and duck at the experimental condition. |
