140 individuals of Longdong White Cashmere Goat and 130 individuals of Zhongwei Goat were selected for analyzing the polymorphism of partial sequence of KAP6.1 and KAP8.1 gene by PCR-SSCP and gene sequencing.In order to analyze the genetic correlation between the polymorphism and the traits of the body weight,and cashmere yeild,the least square linear model was used to determine the preponderant gene locus,the selection reaction was calculated via genetic correlation between partial economic traits and marker loci, providing theoretical basis for MAS in the two goat breeds.The results as follows:1.The analysis results showed that there were polymorphism at the loci of KAP6.1 and KAP8.1 in the two goat breeds.Three genotypes of AA,AB and BB were detected at four loci from six loci in the two breeds.Sequence analysis results showed that there were transition of transversion in 10 places in BB genotype,AA genotype was normal.2.The population genetics analysis showed:.The frequencies of the four loci were analyzed usedλ-test.The result revealed that the Zhongwei Goat population was at Hardy-Weinberg equilibrium at the L1,L2,L3 locus respectively,but was not at Hardy-Weinberg equilibrium at the L4 locus.The Longdong White Cashmere Goat population was at Hardy-Weinberg equilibrium at the L3 locus only,but was not at other three loci.The average effective allele number was 1.5196 which was similar to the observed allele number and this implied that the distributions of the alleles were equilibrium in the two breeds.The PIC and Shannon's information index(S) were much higher in the Longdong White Cashmere Goat population than those in Zhongwei Goat population at the L1,L3 and L4 loci,but lower at L2 locus,and this suggested that the Longdong White Cashmere Goat population contained genetic diversity.At the L1,L3,L4 loci,the PIC was much higher than Zhongwei Goat population,it revealed that genetic diversity was ample in these loci.3.In weight traits,BB genotype was significant difference(P<0.05) with AA genotype at L1 locus,BB genotype was the best marker genotype;AA genotype was significant difference(P<0.05) with BB genotype at L2 and L3 loci,AA genotype was the best marker genotype.Selection reaction showed that L1 locus was the best marker locus as Longdong White Cashmere Goat and Zhongwei Goat in weight trait.4.In cashmere yield,BB genotype was significant difference(P<0.05) with AA and AB genotypes at L1 locus,BB genotype was the best marker genotype.AA genotype was significant difference(P<0.05) with AB genotype and BB genotype at L2 and L3 loci,AA genotype was the best marker genotype.By calculation selection reaction,L2 locus was the best marker site in cashmere yield in the two goat breeds.5.Combining with the genotypes selection,BB genotype at L1 locus could be the preponderant marker locus both for body weight and cashmere yield in Longdong White Cashmere Goat;AB genotype at L3 locus could be the preponderant marker locus both for body weight and cashmere yield in Zhongwei Goat.
