| Pepper is an annual or perennial plant,which belongs to Capsicum genus,Solanaceae family.As one of chinese five major vegetable crops(tomatoes,cucumbers,cabbage,Chinese cabbage,pepper),pepper's fruit is the main nutrition organ for consumption,and its vitamin C content is the highest of vegetables.Also,pepper has high content of beta-carotene, vitamin B1,B2,B5 and contains taste substances,such as capsaicin,dihydro capsaicin, vanilla amine;pigment substances,such as flavin,red-hot pepper element as well as citric acid,tartaric acid,malic acid and other organic acids.China has plenty of pepper resources,and these resources had not been effective in use for a long time,mainly lacked in deep understanding and system excavation.This study used FENGZI PEPPER ONE for tissue culture,and used the material and other 20 materials for genetic diversity to make clear of the genetic basis of these 21 resources at molecular level,laying the foundation for future effective use of a more rational use.1.Establishment of regeneration system on purple pepperDuring bud formation,the best explant type,hormone and sugar constitution for axillary bud formation were terminal bud with 4 leaves and MS+B5(organic elements) +6.0mg/L 6-BA+0.1mg/L NAA+20 g/L sugar respectively;During bud proliferation, hormone constitution of 6-BA with NAA was better than the constitution of 6-BA withIBA,the best hormone constitution for bud proliliferation was 6.0mg/L 6-BA+0.1 mg/L NAA,with the largest proliliferation buds total number and proliferation rate were 145 and 2.42;Medium made large influence on axillary bud rooting,the best hormone constitution was 1/2MS+IBA0.5mg/L,its largest rooting rate were 62.5%;fund influenced plant tranplanting significantly,and when transplanted in Sand:Perlite:Peat soil(1:1:3),the biggest survival rate of cultured plants reached 96.7%.2.The analysis of relationship among some cultivarsHigh quality of DNA obtained from CTAB method,reaching 20-50ng/5ul,and could used for RAPD analysis.Screened from 100 primers,15 primers of 10 bp could amplified 103 distinct band belts,with 82 being polymorphic,and its polymorphism ratio was 79.61%.21 pepper varieties,including FENGZI PEPPER ONE,were identified for their relationship. when the genetic distance was 0.68923,21 varieties could assembled as one large group;when genetic distance reached 0.16854,Wampee sweet pepper and Bianjiao PEPPER ONE were firstly gathered together.when the genetic distance was 0.5122,pepper were divided into five groups.The first group included 14 kinds and had 3 sub-group.The first sub-group included line pepper, long pepper and sweet pepper,and had not formed the clusting similar to morphological classification;FENGZI PEPPER ONE was divided into the third sub-group lonely,and could became one good breeding material because of large difference with other materials on molecular and morphological level;The second group included 3 foreign colorful sweet peppers of JU XIYA,DUO MI and HUANG OUBAO;The third group involved 2 hot clustered-fruit pepper;The fourth and fifth group were QI CUNHONG of Gan Su province and GUI HAIXIANG of Gui Zhou province respectively. |
