In order to understand the infected condition of Crylopora and its epidemio- logical characteristics in Kaifeng of Henan Provice, samples of fresh stool obtained from the patients, who were treating in 2 hospitals of Kaifeng were detected for oocysts of Crylopora by sugar centrifugal flotation method and modified acid-fast stain between June 2007 and May 2008. The results revealed that the total infection rate of Cyclospora was 0.49%(20/4067). (1) The positive rates of Cyclospore cayetanensis infection in age of 0-1,1-5,6-10,1-20,21-30, 31-40,41-50,51-60,61-70, 71-80,8-90, 91-100 were significantly different, with the rates of 0.64 % (1/157) , 0.53% (1/190), 1.10 % (1/91), 0.85% (3/354),0.62%(3/485),0.42%(2/478), 0.75%(4/532), 0.29%(2/696), 0.53 % (3/567),0,0,0 respectively. (2) The infection rates of males and females were 0.36 % (9/2490) and 0.70 % (11/1577) respectively. The experiment of epidemiological investigation of human Cyclospora was carried out in Kaifeng.Ten Cyclospora isolates derived from hospitalized patien were made cross- transmission experiment, nested PCR-RFLP detection and analysis; genotypic analysis of the Small-Subunit rRNA (18S rRNA) gene were carried out to indentify species determine molecular phylogenetic relationship between isolates drived from human of this area and other species, conclude human infection source and transmission mechanism in order to provide theoretical basis for prevention of human crylopora.In order to determine species/ genotype of cyclospora isolates from human in Kaifeng, 18S rRNA specific fragments of isolates were amplified by nested PCR and sequenced. Then, Blast method was used to search homological sequences in NCBI, DDBJ and EMBL, after that, homological sequences were alignmented. Phylogenetic tree and homological analysis were made by some biological softwares such as Clustal X 1.81, and DNAstar 4.0. Based on the phylogenetic analysis of 18S rRNA gene, the ten isolates were idenfied as C. cayetanensis.
