With the rise of the liquid spawn fermentation, cultivatin or extracting substance with biological activity has become the industry hot spot. Liquid strain culture has advantage over the solid strain culture in mechanical efficiency, yield, cultivation, management, etc. Many reports focus on the spawn selection and medium optimization, while the systematic research is deficient and the liquid spawn inoculation device can not meet production's need. Faced with such situation, the pleurotus ostreatus which are cultivated in large scale are discussed. This research concludes strain selection, medium optimization, environment parameter optimization, liquid spawn culture in fermentor and the cultivation application. The automatic inoculation devices are designed. The main conclusions are as follows:1. Five mushroom species are tested to choose the best using liquid culture method. The result turns to be XIAO BAI PING using one way anova. The suitable liquid culture time is 5d.2. The formula of liquid medium is concluded through one factor and orthogonal experiments. the best medium are: sucrose 3%, cotton powder 4%, yeast extract 0.5%, wheat bran 2%, KH2PO4 0.25%, MgSO40.1%, bean oil 0.05%.3. The suitable temperature, pH, and DO are studied through the single-factor and orthogonal experiments, the results are: temperature 25℃, initial pH 6 and 400ml liquid medium in 500ml flask. The suitable fermentation time is 6d. The rotating speed in fermentation tank is controlled in 180rpm. The ventilation volume is controlled in 6L/min. The inoculation quantity is controlled in 10%. The suitable culture day is 3d.4. The liquid spawn were cultivated to study the medium formula. The suitable formula is: cotton seed hull 40%, sawdust 30%, reed 20%, bran 5% and lime 5%.5. Design the hardware that includes microcomputer SST89E516RD, stepper motor 57BYBG and WZM-2H057M , slider cranker mechanism, keyboared Intel 8279 and display circuits OCM4×8, sensors and software systems for inoculation device.This project provides technical support by liquid strain culture, cultivation and inoculation device development, and spread liquid strain more quickly from laboratory to production factory.
