The growth trait of raccoon dog was controlled by multipile genes, the research results of human and mouse showed that GH, GHR, IGF-I and IGF-IR play an regulatory role in growth and development, therefore, the GH, GHR, IGF-I and IGF-IR gene were selected as the candidate gene in our study, and the primer of raccoon dog GH, GHR, IGF-I and IGF-IR gene was designed according to the dog GH, GHR, IGF-I and IGF-IR gene sequence, PCR and sequencing method were developed to clone the exon sequence of GH, GHR, IGF-I and IGF-IR gene. PCR-SSCP method was developed to detect the polymorphism of GH, GHR, IGF-I and IGF-IR gene in the raccoon dog random population, then the association analysis was carried out between the polymorphism and growth traits, and the combined genotype analysis was also carried out, the main results are as follows:1. The CDS sequence of raccoon dog GHR gene was cloned.2. Two SNPs were detected in raccoon dog GHR gene, which were located in exon5 and exon9, the association analysis results between polymorphism and traits showed that the exon9 polymorphism of raccoon dog GHR gene has significant effect on growth traits.3. Two SNPs were detected in raccoon dog IGF-I gene, which were located in exon3 and 3'UTR, the association analysis results between polymorphism and traits showed that the exon3 polymorphism raccoon dog IGF-I gene has significant effect on growth traits.4. The combined genotype analysis result of GHR9 polymorphism site and IGF3 polymorphism site showed that the combined genotype has significant effect on raccoon dog skin length, and the difference between the homozygous combined genotype was much significant than the difference between the homozygous genotype of single polymorphism site.
