Lipoprotein Lipase(LPL)is one kind of key enzyme of animal tissue fat deposition.It catalyzes the hydrolysis of TG and regulates the fatty metabolism.It also plays an important role in facilitating lipoprotein absorption and lipide metabolism and transference by connecting lipoprotein and cell surface protein. So, LPL gene is regarded as the important gene influencing on fat deposition. As fat deposition economy animal, Wild boar and Swine are very different in fat deposition capability, capability of IMF and PUFA.Research on the differential transcription of LPL in Wild boar, Crossbreed pig and Swine will realize regulation in fat deposition and provide more information for Wild boar resources use.We investigated the transcription characteristic of LPL in heart, liver, spleen, lung, kidney, stomach, large intestine, small intestine, longissimus and thigh muscle of Yorkshire, Crossbreed and Wild boar by RT-PCR and detected the concentration of LPL mRNA in longissimus of Yorkshire, Crossbreed and Wild boar by QC-PCR at 3, 6, 9, 12-month old.The results are as follows:1.Results showed that the LPL gene expresses in heart, liver, spleen, lung, kidney, stomach, large intestine, small intestine, longissimus and thigh muscle of Yorkshire, Crossbreed and Wild boar at 3, 6, 9 and 12-month .Although LPL mRNA keeps high expression in heart, low expression in liver and lung.It also expresses in spleen, kidney, stomach, large intestine, small intestine and muscle, but has specificity in space and tissue. It is the highest in musculature of Yorkshire and the lowest in musculature of Wild boar at 12-month.2.The transcription of LPL in Crossbreed's heart was higher than Yorkshire's at every growth stage.Expression regulation of LPL mRNA in liver, spleen, kidney, large intestine, longissimus and thigh muscle of Yorkshire is different from Crossbreed pig on the transcription curve, the growth stage which expression peak is at and expression level. Transcription curve of Yorkshire thigh muscle is different from Crossbreed pig, in the mass expression level ascends in longissimus of Yorkshire and Crossbreed pig at the whole growth phase, but expression peak of LPL mRNA in Yorkshire is at 9-month, which is at 6-month in Crossbreed pig. The concentrations of LPL mRNA in longissimus of Crossbreed pig at 3-month and 6-month separately are as three times and twice as Yorkshire, 4.98E-4μg/ml and 7.88E-4μg/ml more than Yorkshire. The concentrations of LPL mRNA in longissimus of Yorkshire at 9-month and12-month separately are 3.42 E-4μg/ml and 1.48E-4μg/ml more than Crossbreed pig. The difference shows growth period of Crossbreed pig is different from Yorkshire, and fat sediment of Yorkshire is more than Crossbreed pig.3.This research examine studied the concentration of LPL mRNA in longissimus of pig by construction homology competition template to carry on the QC-PCR. The concentrations of LPL mRNA separately are 2.06E-4μg/ml,6.02E-4μg/ml,1.21E-3μg/ml,1.08E-3μg/ml in longissimus of Yorkshire, which separately are 7.02E-4μg/ml,1.39E-3μg/ml,8.68E-4μg/ml,9.32E-4μg/ml in longissimus of Crossbreed pig at 3, 6, 9, 12 -month. The concentration of LPL mRNA is 9.10E-4μg/ml in longissimus of Wild boar at 12-month. Designed primers in the examination of LPL mRNA by QC-PCR only need 33 circulations. And the results could reach the precision for pg/ml. QC-PCR is one economy, quick, accurate quantitative PCR which does not rely on the real-time PCR. Examination result of LPL mRNA in longissimus of Yorkshire and Crossbreed pig by RT-PCR are no significant difference from the result by QC-PCR, corresponding with the laws of the fat sediment in musculature.
