At present in allusion to production of Isoflavones in my country.Discuss on method of using explant tissue culture to product soy isoflavones,to distill Isoflavones,for the future industrial soy isoflavones production is the reason of study. In allusion to soybean,selecting prime explant and prime culture medium of cullus,analysing on wild soybean,selecting content of Isoflavones in leaves of wild soybean,for explant tissue culture to product soy isoflavones lay a foundation.The main results were as followed:(1)Dongnong42 the contents of daidzein and genistein in roots of soybean seedling treated with light for 30 days are 3.69 and 3.59 times of those without treatment. Contents of daidzein and genistein in leaves with light of 30 days are twice of those without treatment. The contents of daidzein and genistein in callus from leaves are highest when cultured in medium for 60 days. The contents of daidzein and genistein in soybean seedlings under lighting condition are higher than those in callus. The total amount of daidzein and genistein in callus of leaves for 90 days are 162.53 and 111.71 times of those in explants. Considering the growth of callus from explants, when callus were cultured for 90 days, root, cotyledon node and cotyledon could be used as explants to produce daidzein and genistein in tissue culture.Hefeng 35 the contents of daidzein and genistein in roots of soybean seedling treated with lighting for 10,20,30 days 2.04 and 1.89 times,2.13 and 1.72 times,1.16 and 1.27 times of those without treatment. The contents of daidzein and genistein in roots of soybean seedling treated with lighting for 20 days are 1.48and1.35 times of those for 10 days, the contents of daidzein and genistein in roots of soybean seedling treated with lighting for 30 days are 1.26 and 1.32 times of those for 10 days. Contents of daidzein and genistein in cotyledons and ctyledonary nodes cultured for 10 days with lighting or not are higher than for 20 days or 30 days.(2)Acupuncturing hypocotyls of soybean seedling and cotyledons explants with Agrobacterium has higher proportion, the ratio is 0.61 and 0.40 times. Way of icision of the lower part of hypocotyls produce hairy root easily, about 35 hairy roots, the contents of daidzein and genistein in hairy roots from central port of hypocotyl are higher. Weight of hairy roots cultured in B5 medium is 2.4 times of those in the other medium.The contents of daidzein and genistein in hairy roots increase in B5 solid medium or B5 liquid medium.But concentrations of daidzein and genistein from hairy roots in B5 liquid medium are 12 and 11 times of those in 1/2MS liquid medium.(3)Biological traits of perennial wild soybean are similar.The contents of daidzein and genistein in leaves from Glycine tabacina are 2.5 and 1.5 times of those from Glycine rabiginosa.But the contents of daidzein and genistein in leaves from Glycine falcata are high relatively,Glycine falcata has big leaves, stem diameter such as biological traits, as high daidzein and genistein contents in the perennial wild soybean resources to research.
