Study On Tissue Culture Technique Of Prunus Campanulata

Prunus spp.is the famous woody appreciates flower.In recent years,the flower cultivation is in imposing array and the seedling wood supply falls short of demand.In this experiment,chose three different provenance of Prunus campanulata from different habitats in Fujian Province as basic material,researched about every steps of the establishment of aseptic system,proliferation,root induction,transplantation etc. Analyzed the key factors of pollution and rootage rate,provided the theoretical basic for the industrial seedling of Prunus campanulata.The main results were described as follows:The aseptic system was established.Sanitized by HgCl₂ or NaCIO and sieved the better method of disinfection is that soak 10 seconds by 70%alcohol,then soak 6 minutes by 0.1%HgCl₂.Then the pollution rate was 36.7%and the survive rate was 45%.Induce the axial bud of Prunus campanulata.Use orthogonal design,then sieved the better scheme of the media for the axial bud induction of P.campanulata FS01 is 1/2MS+4.0mg/L BA 0.025mg/L IBA+1.0g/L AC,and the embryonic rate is 87.5%;the better scheme of the media for the axial bud induction on P.campanulata FS02 is 1/2MS+2.0mg/L BA+0.05mg/L IBA+1.2g/LAC,and the embryonic rate is 84.6%;the better scheme of the media for the axial bud induction on P.campanulata FS03 is 1/2MS+3.0mg/L BA+0.075mg/LIBA+0.8g/LAC or 1/2MS+4.0mg/L BA+0.025mg/L IBA+I.0g/LAC,and the embryonic rate is 80.0%.The proliferation of Prunus campanulata.Use orthogonal design,then sieved the better scheme of the media for the proliferation of P.campanulata FS01 is MS+1.0mg/L BA+0.15mg/L IBA+1.0mg/L GA₃,and the number of proliferation is 5.20 after 30 days; the better scheme of the media for the proliferation of P.campanulata FS02 is MS+1.0mg/L BA+0.03mg/L IBA+0.1mg/L GA3 or MS+1.0mg/L BA+0.08mg/L IBA+1.0mg/L GA₃,and the number of proliferation is 5.00 after 30 days;the better scheme of the media for the proliferation of P.campanulata FS03 is MS+2.0mg/L BA +0.05mg/L IBA+1.0mg/L GA₃,and the number of proliferation is 6.00 after 30 days. The rootage of Prunus campanulata.Use orthogonal design,then sieved the better scheme of the media for the rootage of P.campanulata FS01,FS02,FS03 is 1/2MS+1.0mg/L IBA+1.0mg/L NAA,and the rootage rate is 90.6%,90.0%,91.7%after 30 days.The transplantation of Prunus campanulata.The better transplantation medium is ocher:vermiculite=1:1 and the survive rate is 75%;The better transplantation time is November and the survive rate is 76.7%;The better transplantation environment is the shed with full mist and the survive rate can reach 76.0%.
