Pinus tabulaeformis witch is grow fast, well quality, unable to endure dry and barren, distributed 14 province (region) in north china, is the most useful coniferous tree species. Shaanxi have about 45% natural forest in china , have the big square of Pinus tabulaeformis and genetic diversity. Along with the need of the wood, Random amplified polymorphic DNA (RAPD) markers were used to measure genetic diversity and genetic structure of 95 individuals from 5 natural populations of Pinus tabulaeformis. The main results are as below:(1) Establish a improved method used to extract total DNA from the endosperm of Pinus tabulaeformis can use to RAPD expand(2) A most suitable RAPD system for Pinus tabulaeformis was established: it's 25μL reaction system which contained 10Xbuffer 2.5μl, 2.0 mM Mg2+, 0.15mM dNTPs, 1.5U Tuq DNA polymerase, 2.5μLPrimer and 60ng/μL DNA template.(3) Random amplified polymorphic DNA (RAPD) markers were used to measure genetic diversity and genetic structure of 95 individuals from 5 natural populations of Pinus tabulaeformis. 73 loci were identified from 10 RAPD primers, out of which 64 loci were polymorphic and accounted for 87.67% of the total genetic diversity at species level.(4) Established the genetic similarity coefficients by UPGMA (un-weighted pair group method with arithmetic) analysis. And in 0.35 level the result showed that the 95 samples could be clustered into three groups. |
