The mini-pig is extensive used in biology research as labory animal. However, ordinary mini-pigs carried with the pathogenic microorganisms is not under the well controlled, which has seriously affected the accuracy and reliability of experiment, so it's application was subjected to certain restrictions. So how to clean the microorganisms became a hotspot. The traditional way to culture SPF pig usually adoptes abdominal delivery and sterile midwifer, the former is the most effective one, but the cost is so high, the latter can also obtain clean pigs, but it's difficult to control the pig's delivery time and to avoid the stress to the sow taken by the midwifery personnel. Since embryo transplantation of pig had succeed, it has aroused people's great interest as an new way to culture SPF pig and to control the diseases, and now the abroad have already cultured the SPF herd through ET.In order to cultivate the labory-use SPF mini-pig and enhance the level of our country's labory animal, this study explored the methods of cultivation SPF mini-pig by ET and aseptic midwifery + hand-feeding. In this experiment, We have performed the research about the ET technology to cultivate the SPF mini-pig and establish some foundation, simultaneously we had tried to find out the method for hand-feeding the mini-pig in isolated environment, and got the following conclusions:1. The superovulation efficiencies of PMSG+HCG and PG+PMSG+HCG on WZSP was 65% and 72.5%respectively(P<0.05), but the difference was non-significant between the two superovulation treamentes (P>0.05) of the numbers of average ovulation and average recovery embryos.2.We setted 1000IU, 1500IU, 2000IU and 2500IU four PMSG dose groups in the experiment. The result of 2000IU group was the best, the average numbers of ovulation and recovery embryos were 26.31 and 20.07 respectively, and the superovulation efficiency was 85%, both indexes were significant difference to other groups(P<0.01); the 2500IU group caused oophoritic cyst, but other groups had not found this phenomenon.3. The embryo recovery of surgical procedures stably reached 72.61%, and the donors can be utilized repeatly; the embryo recovery of ex vivo operation was 81.56%, which was significant difference than the former (P<0.01), but the cost was too high.4. The pregnancy rate of embryonic implantation and the rate of embryo survival in this experiment were very low, and got only 30% and 7.57%. The primary reason was the recipients for 4-6 month-old young sows and few sows causing difficults in synchronization of recipients. Moreover, another reason is that the lower embryos'viability may owe to the bad control of temperature.5. Growth indexes had no significant difference between hand-feeding piglets and normal breeding ones before the piglets were raised 20d, but significant difference after raised 20d. The correlation of each growth indexes between hand-feeding piglets and normal breeding piglets was very high, which indicated that the hand-feeding piglets had a slowly growth rate , but it's growth was normal. So it can be used in culturing labory mini-pig.6. The raising way by aseptic midwifery and hand-feeding can clean the Swine Plague, Pseudorabies, PRRS, Swine Flu, Toxoplasmosis, Brucellosis and Chlamydia, although it can't completely eliminate the PPI, it can also be a scientific and effective method for cultivating SPF herd. The effect of this kind of purification needed further advanced research, then we should select the negative ones to be the female parent when this method was adopted to purify herd.7. The diseases including FMD,Brucellosis,Pseudorabies,Swine Plague,PRRS,PCV,Swine Flu,Toxoplasmosis,PPI and Chlamydia can be eliminated completely by operation according to require- ment of the International Embryo Transfer Association.8. This experiment proved it was feasible to cultivate SPF herd by adopting ET technique. It was the first time in our country to cultivate SPF herd by this method, and it also offered us a scientific and effect- ive method to cultivate SPF herd.
