Bread wheat were planted about 1.4 million hectare every year in Sichuan province, and the total production reached 6.0 million ton.But,wheat production was difficult to develop well because of three big problems that were poor quality,low yield and susceptible to stripe rust.To improve quality was one of the most important works in Sichuan province.Because HMW-GS and 1B/1R translocation were important factors for wheat quality,HMW-GS and 1B/1R translocation were analysised on resent released wheat varieties in Sichuan province by SDS-PAGE and PCR.The mayor results as follow:1.The endosperm storage proteins of 44 Sichuan wheat varieties released in 2001-2005 were fractionated by SDS-PAGE to determine their high molecular weight glutenin subunit (HMW-GS) composition.Sixteen pattern of subunits and ten alleles were identified,two alleles at Glu-A1 locus,five alleles at Glu-B1 locus,and three alleles at Glu-D1 locus.The most common pattern ofsubunits were N,7+9,2+12 and 1,20,2+12,their occurrence frequencies were 15.9%and 13.6%,respectively.The glutenin quality scores ranged form 4 to 10,with an average of 6.9,except a novel pattern of subunits(N,20,5+12). 2.36 varieties were amplified by 2 pares PCR primers to test 1B/1R translocation.Result show that 17 varieties contained 1B/1R translocation,and their occurrence frequencies were 47.2%. 3.12 1B/1R translocations from Sichuan varieties were amplified by 5 pares PCR primers on 1RS,results indicated 1B/1R translocations in Sichuan varieties were difference;11 1B/1R translocations from introduction varieties also were amplified by same primers,but the deference only appeared among 3 pares PCR primers,results indicated 1B/1R translocations in introduction varieties also were difference,although 1B/1R translocations in introductions varieties(such as Lovrin,Predgomia,Kavkaz,et al) were original from same Rye variety Petkus(2x).23 varieties were divided into 7 types by PCR using 5 pares PCR primers,result indicated that 1B/1R translocations in varieties were not completely same,they had diversity.
