Wheat backbone parents Aimengniu which contains the synthetically advanced character of dwarf status, multiple disease resistance, high yielding capicity , proper autumn, has high combining and heredity ability as well as many advanced dominant genes, and it was successfully utilized in breeding programs. In this study, homoeologous group 1-specific DNA polymorphic markers were used to reveal genetic difference among the 7 types of Aimengniu, and the transmission of chromosome 1R in 7 types and 34 derivatives of Aimengniu was analyzed by molecular and biochemistry method, which definited the transmission characteristics of 1R. Main results were as follows:(1) One hundred and thirty-eight specific DNA polymorphic markers among 3 parents (Aifeng3, Mengxian 201 and Neuzucht) were selected from homoeologous group 1 DNA specific polymorphic markers, 90 of which were chromosome-specific markers. The results showed that the number of polymorphic alleles between Neuzucht and any of the other 2 parents was more than that between Aifeng3 and Mengxian201, AMNâ… and AMNâ…¢had low-level genetic difference in homoeologous group 1, and any 2 of the other 5 types, AMNâ…¡, AMNâ…£, AMNâ…¤, AMNâ…¥and AMNâ…¦had low-level genetic difference in homoeologous group 1, too. In addition, allelic variation and heterozygous alleles were still detected in 7 types of Aimengniu, which revealed that more allelic variation occured in AMNâ…¦, and heterozygous alleles mainly existed in AMNâ…¡and AMNâ…¦.(2) Chromosome 1B and 1R specific DNA markers were used to detect whether the chromosome 1B and 1R exist or not in 7 types of Aimengniu, at the same time, A-PAGE and multiplex PCR were used to detect the existent of secalin. The results showed that Aimengniuâ… andâ…¢contained the normal chromosome 1B and they did not contain chromosome 1R, while Aimengniuâ…¡,â…£,â…¤,â…¥andâ…¦were 1BL/1RS translocation line and they did not contain 1BS and 1RL.(3) Fifteen 1RS specific DNA markers were used to analyze the transmission of 1R in 34 derivatives, at the same time, A-PAGE and multiplex PCR were used to detect the existent of secalin. The results indicated that 25 derivatives were 1BL/1RS translocation lines, while 8 ones contained the normal chromosome 1B and they did not contain chromosome 1R, and perhaps 1 derivative (Lumai5) contain part segments of 1RS.
