It is one of important wheat breeding work to cultivating wheat Varieties that the high yield and high quality. However, in the past ,the breeding work has made use of minority to inherit resource, and that caused the heediary basis narrow , lead to resistant forfeit , limited the wheat output and the quality potential enhancement. It is the method which using conventional and the molecular breeding to make one of the new germplasm, rich germplasm of resources storehouse most effective way. That is to say, inducing mutafacient mutant, choosing the character beneficial mutant type, to cultivate a high yield and high quality then by that much generation sifting and identifying the queen, and synthesize new good wheat of properties kind of quality resource.This research use different time density, the gradient EMS processing wheat variety HeNong 822, determined the EMS processing wheat variety is suitable the process condition. Discussed M1 ,M2 generation of phenotype variation effect as well as mutation choice method. M2 generation of wheat mutagenic the effect and carries on molecular and the biochemistry examination to the partial mutation.1. Considering death rates and other experimentation factors, we determined 10h and 0.3% are the best process condition in this experiment for HeNong822 treated with the different EMS duration and concentration.2. 7 mutation types were occurred which were preliminary identification according to the phenotype variation, including leaves, stature, panicle department, grain and its physical characteristics. Generally speaking, M1 mutation plants 803, The mutation frequency was 17.6%, in which there are157copies beneficial mutation, accounting for 1.97%. the early flower mutation frequency was the highest, amounted 1.64%. M2 mutation plants 3063, The mutation frequency was 14.6%, in which there are918 copies beneficial mutation, accounting for 29.97%. the dwaring mutation frequency was the highest, amounted 1.75%. M2 mutation the early flower resumption frequency amounted 73.7%; the dwaring mutation amounted 57.5%;big panicle mutation amounted 65.4%3. 3 Compared with the parents, the mutants varied not only on the DNA level, but also on the protein level, which supplied hard evidence for the truth of the Wheat mutant. through SSR examination, partly locus have the variation, the difference in molecular level of the different character were directly reflected by the polymorphism fragments expanded by different primers, through HWM SDS-PAGE examination, the difference of molecular level were indirectly reflected by the different glutenin.
